Cloning and expression of porcine Colony Stimulating Factor-1 (CSF-1) and Colony Stimulating Factor-1 Receptor (CSF-1R) and analysis of the species specificity of stimulation by CSF-1 and Interleukin 34

Abstract: Highlights► The active region of porcine CSF-1 and full length CSF-1R have been cloned. ► Biological activity of porcine CSF-1 has been demonstrated. ► The cross species reactivity of IL-34 has been investigated. ► Both mouse and human IL-34 are biologically active on the pig CSF-1R. ► CSF-1 and IL-34 structure analysis of species-specific activity has been performed.

“…The presence of feline CSF-1R in Ba/F3 cells (Ba/F3fCSF-1R) permitted proliferation in response to rhCSF-1 and removed the dependence of these cells for IL-3. Western blot analysis of these cells demonstrated detectable expression of feline CSF-1R at a similar level to porcine CSF-1R expressed in Ba/F3 cells [2] (Fig. 3).…”

“…2C and D). As noted previously in an analysis of pig CSF-1R [2], it is difficult to predict from these changes whether the cat receptor will bind CSF-1 or IL-34 from other species, and the results may inform both therapeutic applications and structure–function predictions.…”

“…Intriguingly, the X. laevis IL-34-derived macrophages possess elevated arginase-1 gene expression and yet also exhibit potent respiratory burst responses, typical of mammalian M1 macrophages. It is notable that IL-34 is found across all vertebrates (Garceau et al, 2010; Gow et al, 2012; Gow et al, 2013; Wang et al, 2013). In fact, this cytokine may even predate CSF-1, considering the presence of IL-34 and absence of CSF-1 in the elasmobranch genome (Venkatesh et al, 2014).…”
