Clofazimine, but Not Isoniazid or Rifampicin, Augments Platelet Activation in vitro

Abstract: Although the inclusion of the cationic amphiphilic, anti-mycobacterial agent, clofazimine, in the chemotherapeutic regimens of patients with multidrug-resistant tuberculosis (TB) has contributed to improved outcomes, concerns remain about the cardiotoxic potential of this agent. Accordingly, the current study was undertaken with the primary objective of investigating the effects of clofazimine, on the reactivity of human platelets in vitro, a seemingly unexplored, mechanism of cardiotoxicity. Platelet-rich pla… Show more

“…After 15 min of incubation in the dark, the samples were analyzed on a Gallios flow cytometer (Beckman Coulter) and the results expressed as the percentage of activated platelets with 50,000 cells interrogated during each measurement. The gating and analytical strategies used in a typical experiment have been described elsewhere [54].…”

“…Salbutamol inhibited the ROS production in RAW264.7 macrophages stimulated by lipopolysaccharide (26) . In neutrophils stimulated by N-formyl-L-methionyl-L-leucyl-L-phenylalanine (fMLP), salbutamol reduced the generation of ROS (27) . Our results are in agreement with the previous studies: salbutamol reduced the ROS production when compared to CSE-stimulated BEAS-2B cells.…”

“…Clofazimine upregulated Cluster Of Differentiation 62 Platelet (CD62P) expression, enhanced ADP, and activation of thrombin-mediated human platelet in vitro , which appears to be a novel cardiotoxic mechanism ( Anderson et al., 2018 ). CD62P is widely recognized as a critical factor in activating endothelial cells and react situations of living blood platelet.…”
