CD151 is a member of the tetraspanin family, which is involved in diverse cellular processes, including proliferation, motility, and invasion. However, the role of CD151 in breast cancer especially luminal and basal-like subtype breast cancer remains obscure. Here, we report the role of CD151 in the biological behaviors of luminal and basal-like subtype cell lines and the underlying molecular mechanism. A eukaryotic expression vector expressing both CD151 shRNA and GFP was transfected into MCF-7 and MDA-MB-468 cells. The CD151 gene-silencing effect is authenticated by real-time PCR and Western blot. Our data show that the capacity for proliferation, migration, and invasion of two kinds of cells is diminished after Knockdown of CD151 via lentivirus-mediated CD151 specific shRNA. Tumor cells are arrested in G0/G1 phase. Apoptosis is increased. Moreover, we also demonstrate that the expressions of mmp26 and CD147 are inhibited by knockdown of CD151. But the inhibition depends on the cell type. We can conclude that silencing gene CD151 inhibits expression of properties that are associated with the malignant phenotype of MCF-7 and MDA-MB-468 cells. It may become a potential target in breast cancer therapy especially for luminal and basal subtypes.

Nowadays, breast cancer has been known as heterogenous disease with various biological features which require different therapy strategies. Breast cancers with lack or no expression of estrogen (ER), progesterone receptors (PR), and human epidermal growth factor receptor 2 (HER-2) are defined as triple negative breast cancer (TNBC). To date, Ki-67 has been used in order to characterize cell proliferation. This study is to analyze Ki-67 labeling index (LI) expression based on clinicopathological features of TNBC. This descriptive study with cross-sectional design was performed. Data about demographics were extracted from patients' medical record and histopathologic feature and immunohistochemistry was done. Results of analysis data were presented in frequency tables. Mean and standard deviation Ki-67 LI were also obtained. In this study, most patients aged 35-49 years old with mean of 46,8 years old. Most of them had bigger tumour size, had already metastasized to lymph node, had no distant metastases and higher stage. Ki-67 LI value in this study quite high with mean of 43,89%. %. If TNBC patients were young, had distant metastases, higher mitotic count (≥15/10 HPF) and higher grade, they tended to have higher Ki-67 LI. Patients with metaplastic carcinoma of no special type and carcinoma with medullary features also tended to have higher Ki-67 LI. Meanwhile tumour size, metastases to lymph node and clinical stage were not in accordance with Ki-67 LI.

Background: The aim of this study was to compare the clinicopathological characteristics of Triple Negative Breast Carcinomas with Non Triple Negative Breast Carcinomas (NTNBC). Evaluation of expression of Epidermal Growth Factor Receptor (EGFR), CK5/6 in TNBC and their comparison with NTNBC was done.Methods: 25 TNBC and 35 NTNBC were selected. The clinicopathological parameters of these two groups were compared. Each group was further immunostained for basal markers(CK5/6 and EGFR).The expression of markers in these two groups was studied and compared with each other. Results:The mean age of TNBC and NTNBC were 47 and 49 years respectively. The majority (48%) of cases from TNBC as well as NTNBC (45%)were in size range of 2-5 cm(T2 stage). IDC-NOS was predominant histological type seen in 92% of TNBC and 100% NTNBC.TNBCs had a significantly higher tumor grade than NTNBC at presentation. LVI was seen in 40% TNBC cases and 42% NTNBC cases. Majority (52%) of TNBC cases were node negative while majority (37.14%) of NTNBC cases belonged to N1 stage. IIA was the most common stage in 36% TNBC cases . In NTNBC, majority of the cases (34%) belonged to Stage IIIA.Expression of basal markers was significantly associated with triple negative breast cancers. Conclusion:TNBCs had a significantly higher tumor grade than NTNBCs at presentation. Expression of basal markers was significantly associated with TNBCs. Since EGFR was significantly associated with triple negative phenotype, TNBC could potentially benefit from EGFR targeted therapeutic strategies.
