Background Exosomes are a promising tool in disease detection because they are noninvasive, cost-effective, sensitive and stable in body fluids. MicroRNAs (miRNAs) are the main exosomal component and participate in tumor development. However, the exosomal miRNA profile among Asian melanoma patients remains unclear. Methods Exosomal miRNAs from the plasma of melanoma patients (n = 20) and healthy individuals (n = 20) were isolated and subjected to small RNA sequencing. Real-time PCR was performed to identify the differential miRNAs and to determine the diagnostic efficiency. Proliferation, scratch and Transwell assays were performed to detect the biological behavior of melanoma cells. Results Exosomal miRNA profiling revealed decreased miR-1180-3p expression as a potential diagnostic marker of melanoma. The validation group of melanoma patients (n = 28) and controls (n = 28) confirmed the diagnostic efficiency of miR-1180-3p. The level of miR-1180-3p in melanoma cells was lower than that in melanocytes. Accordingly, the level of miR-1180-3p was negatively associated with the proliferation, migration and invasion of melanoma cells. Functional analysis and target gene prediction found that ST3GAL4 was a potential target and highly expressed in melanoma tissues and was negatively regulated by miR-1180-3p. Knockdown of ST3GAL4 hindered the malignant phenotype of melanoma cells. Conclusions This study indicates that reduced exosomal miR-1180-3p in melanoma patient plasma is a promising diagnostic marker and provides novel insight into melanoma development.

Background: The immune microenvironment of lung cancer is complex, and T-cell tolerance is a primary mechanism of immune evasion. Tumor-derived exosomes (TEX) miRNAs regulate immune cells in tumor microenvironment (TME), contributing to tumor immune suppression. However, the underlying mechanisms remain unclear.Methods: TEX miRNA expression dataset (GSE111803) and CD8+ T cells mRNA expression profile (GSE90728) in lung adenocarcinoma (LUAD) were downloaded from Gene Expression Omnibus (GEO). The differentially expressed miRNAs (DEMs) and differentially expressed genes (DEGs) were analyzed. The intersection of target genes of DEMs and DEGs was defined as the overlapped target genes. We performed the function and pathway analysis for DEGs and the overlapped genes by the DAVID tool and constructed a protein-protein interaction (PPI) network to screen modules and hub genes. Results: In total, 7 DEMs (5 upregulated and two downregulated miRNAs) and 672 DEGs (219 upregulated and 453 downregulated genes) were identified, then 185 overlapped genes for upregulated DEMs were selected for further analysis. GO analysis indicated the DEGs were mainly enriched in positive regulation of B cell activation, innate immune, while the overlapped genes were primarily involved in regulation of immune response. The pathway analysis suggested DEGs were primarily enriched in cytokine-cytokine receptor interaction, while the overlapped genes were primarily enriched in osteoclast differentiation. The PPI network revealed three significant modules for DEGs and the overlapped genes sets. These modules genes were mainly enriched in cell cycle, chemokine signaling pathway, and cytokine-cytokine receptor interaction, respectively.Conclusions: These results preliminarily revealed the mRNA expression profiles and the molecular regulatory mechanism of TEX miRNA regulated tumor immune response in LUAD.

An early diagnosis and effective prognostic factors would greatly reduce the mortality rate of colorectal cancer (CRC). This research is intended to complete the evaluation of the prognostic value and potential role of miR-1180-3p in CRC. The miR-1180-3p levels were reduced in CRC patients’ tissues, blood, and human CRC cell lines. The ability of miR-1180-3p was explored in discrimination of CRC patients and healths and the value in overall survival estimate. The effect of miR-1180-3p dysregulation on the CRC cellular function was investigated. miR-1180-3p is downregulated in CRC tissues, blood and cells than normal ones. This lower expression was correlated with vascular invasion, lymph node metastasis, and TNM stage. With the use of ROC curve, miR-1180-3p showed discriminating ability in CRC patients and healthy subjects. With the result of Kaplan–Meier analysis and multi-multivariate Cox analysis, miR-1180-3p was an independent predictor for CRC patients’ overall survival. Utilizing CCK-8, Transwell and matrigel assays, overexpression of miR-1180-3p reduced cancer cell proliferation and mobility, but induced apoptosis, by targeting COL12A1 . miR-1180-3p might function as a suppressor in CRC progression and allowed the discovery of a new biomarker for diagnosis, prognosis and therapy target for CRC.
