Clinical Performance of the PreTect HPV-Proofer E6/E7 mRNA Assay in Comparison with That of the Hybrid Capture 2 Test for Identification of Women at Risk of Cervical Cancer

Abstract: Human papillomavirus (HPV) DNA testing has a higher clinical sensitivity than cytology for the detection of high-grade cervical intraepithelial neoplasia or worse (CIN 2؉). However, an improvement in specificity would be desirable. As malignant transformation is induced by HPV E6/E7 oncogenes, detection of E6/E7 oncogene activity may improve specificity and be more predictive of cervical cancer risk. The PreTect HPVProofer assay (Proofer; Norchip) detects E6/E7 mRNA transcripts from HPV types 16, 18, 31, 33, a… Show more

“…According to the present results, the application of E6/E7 protein detection in CC screening could reduce the limitations of other recent technologies, such as the Pap test and HC2 test, and the indicators (E6/E7 proteins) for both cervical pre-cancer and progression to CC could be quantified with the standard detection method established in the present study (17,21). However, not all types of anti-E6/E7 monoclonal antibodies exhibit high specificity currently (8,38).…”

“…Histology is recognized as the gold standard for diagnosing the pathological process of CC. CIN2 + is considered a precancerous lesion and, if left untreated, this may progress to cervical cancer; therefore, if a patient exhibits a CIN2 + lesion, this should be treated (17,29). However, it is unknown which kind of lesions will finally lead to infiltrative cancers.…”

“…Commercially available robust assays for HPV mRNA detection can be performed in reflex after liquid based cytology or the HPV DNA test (4,9,22,26,28,30,33,38). We compared here the clinical performance of the PreTect HPVProofer E6/E7 mRNA assay (Norchip, Klokkarstua, Norway) as a triage test to that of the HPV DNA test and cytology for detecting high-grade cervical intraepithelial neoplasia or worse (CIN2ϩ).…”
