Clinical Implications of TMPRSS2-ERG Gene Fusion Expression in Patients With Prostate Cancer Treated With Radical Prostatectomy

Abstract: TMPRSS2-ERG fusion gene status classifies patients with prostate cancer treated with radical prostatectomy into groups defined by different prognostic factors. This could be the basis for designing more refined treatment strategies.

“…We found no association of ERG rearrangements with cancerspecific survival. The prognostic value of the TMPRSS2-ERG fusion gene is controversial in the literature when using the same end point described here (Demichelis et al, 2007;Fitzgerald et al, 2008;Rubio-Briones et al, 2010), but also among authors using BR as end point (Perner et al, 2006;Lapointe et al, 2007;Mehra et al, 2007;Nam et al, 2007;Saramäki et al, 2008;Yoshimoto et al, 2008). The last large FISH-based report to evaluate the clinical relevance of the TMPRSS2-ERG fusion in patients treated with RP found no association of the fusion gene with clinical outcome (Gopalan et al, 2009).…”

“…These data, together with our latest results, confirm the potential role of SPOP as tumour suppressor in PCa. T2E has already been described to define a subgroup of PCa tumours with different clinico-pathological characteristics and prognostic behaviour [14]. Moreover, certain lesions have been found to be mutually exclusive with the presence or absence of ETS rearrangements [15].…”

“…Several nomograms combining PSA with other clinical variables have also been developed to improve prognostic value beyond that of individual tests 16 Clinical significance of these PSA derivatives has been reported elsewhere. Serum PSA circulates in either an unbound "free" form or bound to one of several proteins, most frequently alpha-1-antichymotrypsin (ACT) 17 . Levels of free PSA (fPSA) can be detected and compared to total PSA, yielding the proportion of free PSA (%fPSA).…”
