Claudin-6 is a single prognostic marker and functions as a tumor-promoting gene in a subgroup of intestinal type gastric cancer

Abstract: Background We aimed to identify novel tumor-promoting drivers highly expressed in gastric cancer (GC) that contribute to worsened prognosis in affected patients. Methods Genes whose expression was increased and correlated with worse prognosis in GC were screened using datasets from the Cancer Genome Atlas and Gene Expression Omnibus. We examined Claudin-6 (CLDN6) immunoreactivity in GC tissues and the effect of CLDN6 on cellular functions in GC cell lines. The mechanisms underlying GC-promoting function of CLD… Show more

“…one report indicated that the expression of cldn6 differed between gastric cancer and adjacent tissues (36). additional studies showed that cldn6 expression differed in gastric cancer compared with healthy tissue (37)(38)(39)(40)(41), and according to Zavala-Zendejas et al (37), the expression of cldn6 in gastric cancer tissues was higher compared with that in adjacent tissues. cldn6 is a gene promoter in gastric cancer, and its upregulation enhances the tumorigenicity of gastric cancer cells (37).…”

“…As for the genomic analysis, 40% of diffused-type GC was associated with CHD1 (chromodomain-helicase DNA-binding protein 1) loss; additionally, diffused-type GC was inclined to have mutant p53 and oncoprotein Her2, which induced diffused-type GC to be more aggressive type ( 13 ). However, other genes that were overexpressed in intestinal-type GC were also associated with poorer survival such as Claudin 6 (CLDN6) ( 14 ). There was little literature to investigate the difference of prognosis for patients with intestinal-type and diffused-type GC.…”

“…CLDN6 had been demonstrated abnormal expression and can be a prognostic marker in cancers including ovarian cancer ( Wang et al, 2013 ), endometrial cancer ( Kojima et al, 2020 ), gastric cancer ( Kohmoto et al, 2020 ), breast carcinoma ( Liu et al, 2016 ; Jia et al, 2019 ), and lung cancer ( Micke et al, 2014 ). Bioinformatic analysis has revealed that CLDN6 is regulated by a diverse set of transcription factors and promotes cancer cell behavior via the ASK1-p38/JNK MAPK secretory signaling pathway ( Lin et al, 2017 ).…”
