Cisplatin Loaded Multiwalled Carbon Nanotubes Induce Resistance in Triple Negative Breast Cancer Cells

Abstract: In this paper we developed a method for multiwalled carbon nanotubes (MWCNTs) use as carriers for a drug based on platinum in breast cancer therapy. The method of functionalization involves the carboxyl functionalization of nanotubes and encapsulation of cisplatin (CDDP) into MWCNTs. The biological properties of MWCNTs loaded with CDDP (MWCNT-COOH-CDDP) and of individual components MWCNT-COOH and free CDDP were evaluated on MDA-MB-231 cells. Various concentrations of CDDP (0.316–2.52 µg/mL) and MWCNTs (0.5–4 µ… Show more

“…Applying such carriers for CP delivery may exert reverse effect and promote drug resistance of cancer cells. It has been reported that CP-loaded multiwalled carbon nanotubes significantly diminish ROS levels and induce failure of CP in mediating apoptosis in breast cancer cells, leading to development of drug resistance [328]. Therefore, this aspect should be considered while synthesizing nanocarriers for CP delivery and suppressing cancer progression.…”

“…The free carboxyl group in the structure of FA-2-DG can be used for drug loading [35]. Cisplatin, a first-line antineoplastic drug with weak water solubility, is often studied to improve its bioavailability, and many nanodrug-loading systems have been developed to achieve this goal [32,36,37]. These studies have shown that forming coordination bonds between carriers and cisplatin is a very effective strategy.…”

“…In recent times, CNTs are widely used as the delivery system of drugs, nucleic acids, proteins, peptides, etc. In addition, they can be easily modified by surface functionalization in order to provide tunable characteristics like low toxicity and non-immunogenicity [ 274 ]. It was further observed that besides enhancing the therapeutic activity of carbon nanotubes, the surface functionalization also increases their aqueous solubility, biocompatibility, and biodegradability, which were considered the drawback of CNTs [ 275 ].…”
