Cisplatin administration influences on toxic and non-essential element metabolism in rats

Abstract: Nowadays several papers deal with the effectiveness and side effects of metal complexes, especially cisplatin, in cancer therapy. The excretion of essential metal elements from the body is a serious problem in the treatment, but there are no data concerning the distribution and metabolism of toxic and nonessential elements. Therefore our aim was to study the concentration of some of these elements after treatment with cisplatin. Male Wistar rats (n=20, 175-190 g) were randomly divided into 2 groups (n=10/group… Show more

“…Wet liver homogenate samples (0.5 g) were digested in the mixture of 3 ml HNO 3 (65%) and 2 ml H 2 O 2 (30%) in an open block digestion system with 200 8C heating, then it was transferred to a 10 ml volumetric flask and filled up with high purity water (18.5 MΩ). Measurements were carried out for Al, B, Ba, Ca, Co, Cr, Cu, Fe, Li, Mg, Mn, Mo, Na, Ni, P, Pb, Si, Sn, Sr, and Zn with a Spectro Genesis simultaneous ICP-OES spectrometer (Spectro Analytic Instruments GmbH, Kleve, Germany) (Szentmihályi et al, 2014). Because of the use of isotonic KCl solution for the preparation, potassium concentration could not be measured.…”

“…A májmintákból 500-500 mg-ot 5 ml 65%-os salétromsav és 2 ml H 2 O 2 elegyével mikrohullámú feltáróban roncsoltunk. Az elemtartalmak mérésére SPECTRO GENESIS ICP-OES (SPECTRO, Kleve, Németország) és iCAP Q ICP-MS (ThermoFisher Scientific, Waltham, MA, USA) készüléket használtunk multielemes standardok (MU33 multielement standard, 100 mg/l, CPA-Chem Product, Stara Zagora, Bulgária és EPA30 multielement standard, 5-100 mg/l, Bernd Kraft GmbH, Duisburg, Németország) segítségével [27].…”
