Circular RNA microarray expression profile in 3,4‐benzopyrene/angiotensin II‐induced abdominal aortic aneurysm in mice

Abstract: Abdominal aortic aneurysm (AAA) is an unpredictable but lethal disease that poses a therapeutic dilemma. Circular RNAs (circRNAs), whose functional roles as transcriptional regulators and microRNA (miRNA) sponges have been shown in former studies, are potential biomarkers for many diseases. AAA in male C57BL/6 J mice was induced by coadministration of angiotensin II (Ang II) and 3,4-benzopyrene (BaP). The circRNA expression profiling was performed using two samples from the control group and two samples from t… Show more

“…24 In AAA, circRNAs expression profiling indicated the potential participation of circRNAs in AAA. 18 Consistent with previous work, we first demonstrated that circRNACCDC66 was elevated in Angiotensin II-stimulated VSMCs, which is prerequisite for probing its function in AAA. As documented previously, circRNACCDC66 was tumour-facilitating in gastric cancer, 25 colon cancer, 26 and lung adenocarcinoma.…”

“…Circular RNA CCDC66 (circCCDC66) was an upregulated circRNA in AAA from previous revelation. 18 Consequently, we probed the function of circCCDC66 in VSMC apoptosis and proliferation via multiple assays in this study. Besides, we explored whether circCCDC66 could modulate its host gene to function in AAA.…”

“…Overexpression of circ_104916 inhibits migration and invasion of colorectal cancer cells via repressing epithelial to mesenchymal transition (EMT) [36]. Wang J et al discovered six circRNAs related to apoptosis, which induced abdominal aortic aneurysm via promoting endothelial cells growth from circRNA-miRNA networks [37]. Circ_0005015 facilitates retinal endothelial angiogenesis by regulating proliferation, migration and tube formation of endothelial cells [38].…”

“…Previously, Zheng et al found that hsa_circ_000595 was upregulated in human AAA tissues, which would reduce the expression of miR-19a and subsequently promote human vascular smooth muscle cells (VSMC) apoptosis [31]. Similarly, apoptosis-related circRNAs were observed altered in AAA animal models [26]. Besides, circWDR77/ miR-124/FGF-2 and circSATB2/miR-939/STIM1 regulatory axis are verified to regulate VSMC proliferation and migration [32,33].…”

“…Using gene expression profile, several researchers detected that immune and inflammatory response had a great impact on the pathogenesis of AAA [22][23][24]. Recently, advances in novel computational approaches and high-throughput sequencing techniques have sparked new interest in the research on noncoding RNAs [25,26]. As an important member of noncoding RNAs, cir-cRNAs are abundant, stable and highly conserved.…”
