Circadian Regulation of Tshb Gene Expression by Rev-Erbα (NR1D1) and Nuclear Corepressor 1 (NCOR1)

Abstract: Background: Basal metabolic rate is regulated by thyroid hormone; the mechanism is unknown. Results: NCOR1 and Rev-Erb␣ enrich at different sites from thyroid hormone receptor on the Tshb promoter. Conclusion: NCOR1 and Rev-Erb␣ interact to regulate circadian expression of Tshb mRNA independent of thyroid hormone. Significance: This novel role of Rev-Erb␣ in Tshb expression reveals new links between circadian rhythms and metabolism.

“…55 In vitro data also indicate that N-CoR targets NR1D1 (also known as Rev-Erbα) not THR at the Tshb promoter to regulate basal circadian expression patterns of this gene. 56 It is also possible that a completely different set of cofactors is recruited when THR is bound to a negative TRE. For example, nuclear factor RIP140 interacts with THRα–RXR or THRβ–RXR heterodimers, and this interaction might lead to inhibition of transcription mediated by THR homodimers.…”

“…The mechanism of circadian TSH secretion is becoming increasingly clear, with the nuclear corepressor, NCOR, now known to impinge on thyroid hormone receptors bound to TSH subumit genes [12]. The 24-h mean concentrations of prolactin in plasma were the same in all groups, whereas those of LH and FSH were twice as high in the elderly as in the young men and eight and 23 times higher, respectively, in the elderly women.…”

“…Therefore, NR1D1 seems to be the principal metronome of the body. NR1D1 regulates the transcription of “the long-day gene” TSHB (thyroid stimulating hormone, beta), and through this action NR1D1 is also a link between the effects of light and the seasonal variation in behavior ( 36 ). Transcription of TSHB is induced to a greater extent about 14 h after dawn of the first long day in the spring by the increasing exposure to light ( 37 ).…”
