The Chinese herbal medicine, Huzhen Tongfeng Formula (HZTF), derived from traditional Chinese medicine (TCM) practice, has recognized therapeutic benefits for gouty arthritis (GA). HZTF is currently in the late stage of approval process as a new anti-GA drug application. However, the underlying mechanism of HZTF as an antigout medication is unclear. In this study, we combined network pharmacology and experimental validation approaches to elucidate the mechanism of action of HZTF. First, the relative drug-disease target networks were constructed and analyzed for pathway enrichment. Potential pathways were then validated by in vitro and in vivo experiments. We found that 34 compounds from HZTF matched 181 potential drug targets. Topology analysis revealed 77 core targets of HZTF, which were highly related to gout, following screening of KEGG pathway enrichment. Further analysis demonstrated that the arachidonic acid metabolic pathway was the most relevant pathway involved in the mechanism of HZTF. Validation experiments showed that HZTF significantly inhibited the inflammatory cell infiltration into gouty joints, improved the swelling of affected joints, and increased the pain threshold. HZTF significantly reduced the transcription and production of various cytokines and inflammatory mediators in vitro. In particular, cyclooxygenase (COX)-1, COX-2, and 5-lipoxygenase were simultaneously downregulated. In conclusion, our study suggests that the antigout mechanism of HZTF is associated with the inhibition of the arachidonic acid pathway, resulting in the suppression of inflammatory cytokines and mediators. These findings extend our understanding of the pharmacological action of HZTF, rationalizing the application HZTF as an effective herbal therapy for GA.

Natural bioactive compounds that trigger apoptosis in medicinal plants might be a promising new source of anti-cancer medications. The present study aimed to assess the anti-cancer potential of the powerful therapeutic plant Chrysanthemum indicum. The whole plant extract was extracted in n-hexane and ethanol solvents to test anticancer and antioxidative capabilities on the cancer HepG2 cell line. The cells in the untreated group had not been exposed to any of the plant extracts. Ethanol and n-hexane extracts were administered to the treated cell lines. Cell viability was estimated by employing standard methodologies for the anticancer potential. Cell death and viability were measured by MTT, trypan blue, and crystal violet tests. The rate of apoptosis was measured in all groups using an Annexin-V ELISA. Furthermore, antioxidant enzyme activity studies were carried out to investigate the scavenging activities of nitric oxide, DPHH radicals, H2O2, and superoxide, scavenging power of the extracts. When cancer cells from the HepG2 cell lines were treated with plant extracts, they showed lower viability, decreased proliferation, and increased apoptosis compared to untreated control cells, according to the findings. Plant extract-treated cancer cells have stronger anti-oxidative scavenging activity than untreated cancer cells. In comparison to the n-hexane extract, the ethanol extract exhibited stronger anti-cancer and antioxidative capability against cancer cells. It was concluded that Chrysanthemum indicum ethanol and n-hexane extracts induce anticancer activity in cancer cell lines by increasing apoptosis, antioxidant status, and decrease proliferation.

Gout is the most common form of inflammatory arthritis. It occurs when monosodium urate crystals (MSU) are deposited within joints due to hyperuricemia and persistent elevations of serum uric acid levels. Traditional gout treatment such as urate-lowering therapy is difficult to continue for a long period of time due to the risk of side effects. Recent studies have shown that the modulation of MSU-induced inflammatory responses is dependent on the inflammatory cytokine IL-1β, which has a central role in a chain of processes involving multiple cytokines and mediators. In this regard, the NLRP3 inflammasome is known to play a crucial part and thus has been proposed as a novel target in the treatment for gout. However, the biochemical mechanism for NLRP3 inflammasome activation has not yet been clearly elucidated. Therefore, this report can provide an overview of natural extractions targeted to prevent or treat NLRP3 inflammasome-mediated gout in the MSU-induced gout model. In addition, the research and development of such natural products are suggested as a potential strategy in the treatment of gout.
