Diatom diversity in lakes of northwest Yakutia (Siberia) was investigated by microscopic and genetic analysis of surface and cored lake sediments, to evaluate the use of sedimentary DNA for paleolimnological diatom studies and to identify obscure genetic diversity that cannot be detected by microscopic methods. Two short (76 and 73 bp) and one longer (577 bp) fragments of the ribulose 1,5-bisphosphate carboxylase/oxygenase (rbcL) gene, encoding the large subunit of the rbcL, were used as genetic markers. Diverse morphological assemblages of diatoms, dominated by small benthic fragilarioid taxa, were retrieved from the sediments of each lake. These minute fragilarioid taxa were examined by scanning electron microscopy, revealing diverse morphotypes in Staurosira and Staurosirella from the different lakes. Genetic analyses indicated a dominance of haplotypes that were assigned to fragilarioid taxa and less genetic diversity in other diatom taxa. The long rbcL_577 amplicon identified considerable diversification among haplotypes clustering within the Staurosira/Staurosirella genera, revealing 19 different haplotypes whose spatial distribution appears to be primarily related to the latitude of the lakes, which corresponds to a vegetation and climate gradient. ABSTRACT. Sub-fossil sections from a Florida wetland were accelerator mass spectrometry (AMS) dated and the sedimentological conditions were determined. 14 C data were calibrated using a combined wiggle-match and 14 C bomb-pulse approach. Repeatable results were obtained providing accurate peat chronologies for the last 130 calendar yr. Assessment of the different errors involved led to age models with 3-5 yr precision. This allows direct calibration of paleoenvironmental proxies with meteorological data. The time frame in which 14 C dating is commonly applied can possibly be extended to include the 20th century.

Archived data and sedimentary macrofossil records of vegetation and invertebrates deposited in a 60-cm long sediment core were analysed to examine if macrophyte dominance was a permanent feature of the recent history of lowland Lake Kleszczów. For the last several centuries, the lake has not been dominated by phytoplankton but by floating-leaved vegetation at strongly reduced water level. Starting from the midnineteenth century, probably as a result of climate fluctuations, vegetation switched at first into submerged angiosperms, and then, in the second half of twentieth century, towards charophytes. Within charophytes there were switches between Chara globularis and C. vulgaris communities, depending on lake productivity or hydrological stress. No symptoms were detected of a switch to a turbid regime as a result of potential internal supply of phosphorus from sediments covered with a dense carpet of charophytes. Our study shows that within a longer period with clear water, the community of macro-vegetation can be highly dynamic. It can be represented by various types of vegetation as a response to different productivity levels and/or hydrological stress, largely determining the composition of other hydrobionts and course of various processes, and as a consequence, the functioning of the ecosystem and its resilience.
