Reliable information on past and present vegetation is important to project future changes, especially for rapidly transitioning areas such as the boreal treeline. To study past vegetation, pollen analysis is common, while current vegetation is usually assessed by field surveys. Application of detailed sedimentary DNA (sedDNA) records has the potential to enhance our understanding of vegetation changes, but studies systematically investigating the power of this proxy are rare to date. This study compares sedDNA metabarcoding and pollen records from surface sediments of 31 lakes along a north-south gradient of increasing forest cover in northern Siberia (Taymyr peninsula) with data from field surveys in the surroundings of the lakes. sedDNA metabarcoding recorded 114 plant taxa, about half of them to species level, while pollen analyses identified 43 taxa, both exceeding the 31 taxa found by vegetation field surveys. Increasing Larix percentages from north to south were consistently recorded by all three methods and principal component analyses based on percentage data of vegetation surveys and DNA sequences separated tundra from forested sites. Comparisons of the ordinations using procrustes and protest analyses show a significant fit among all compared pairs of records. Despite similarities of sedDNA and pollen records, certain idiosyncrasies, such as high percentages of Alnus and Betula in all pollen and high percentages of Salix in all sedDNA spectra, are observable. Our results from the tundra to single-tree tundra transition zone show that sedDNA analyses perform better than pollen in recording site-specific richness (i.e., presence/absence of taxa in the vicinity of the lake) and perform as well as pollen in tracing vegetation composition.

We investigated the subfossil chironomid fauna of 150 lakes situated in Yakutia, northeastern Russia. The objective of this study was to assess the relationship between chironomid assemblage composition and the environment and to develop chironomid inference models for quantifying past regional climate and environmental changes in this poorly investigated area of northern Russia. The environmental data and sediment samples for chironomid analysis were collected in 5 consecutive years, [2003][2004][2005][2006][2007], from several regions of Yakutia. The lakes spanned wide latitudinal and longitudinal ranges and were distributed through several environmental zones (arctic tundra, typical tundra, steppetundra, boreal coniferous forest), but all were situated within the zone of continuous permafrost. Mean July temperature (T July ) varied from 3.4°C in the Laptev Sea region to 18.8°C in central Yakutia near Yakutsk. Water depth (WD) varied from 0.1 to 17.1 m. T July and WD were identified as the strongest predictor variables explaining the chironomid communitiy composition and distribution of the taxa in our data set. Quantitative transfer functions were developed using two unimodal regression calibration techniques: simple weighted averaging (WA) and weighted averaging partial least squares (WA-PLS). The two-component T July WA-PLS model had the best performance. It produced a strong coefficient of determination (r 2 boot = 0.87), root mean square error of prediction (RMSEP = 1.93), and max bias (max bias boot = 2.17). For WD, the one-component WA-PLS model had the best performance (r 2 boot = 0.62, RMSEP = 0.35, max bias boot = 0.47).

Changes in species’ distributions are classically projected based on their climate envelopes. For Siberian forests, which have a tremendous significance for vegetation-climate feedbacks, this implies future shifts of each of the forest-forming larch (Larix) species to the north-east. However, in addition to abiotic factors, reliable projections must assess the role of historical biogeography and biotic interactions. Here, we use sedimentary ancient DNA and individual-based modelling to investigate the distribution of larch species and mitochondrial haplotypes through space and time across the treeline ecotone on the southern Taymyr peninsula, which at the same time presents a boundary area of two larch species. We find spatial and temporal patterns, which suggest that forest density is the most influential driver determining the precise distribution of species and mitochondrial haplotypes. This suggests a strong influence of competition on the species’ range shifts. These findings imply possible climate change outcomes that are directly opposed to projections based purely on climate envelopes. Investigations of such fine-scale processes of biodiversity change through time are possible using paleoenvironmental DNA, which is available much more readily than visible fossils and can provide information at a level of resolution that is not reached in classical palaeoecology.

a b s t r a c tA 380 cm long sediment core from Lake Temje (central Yakutia, Eastern Siberia) was studied to infer Holocene palaeoenvironmental change in the extreme periglacial setting of eastern Siberia during the last 10,000 years. Data on sediment composition were used to characterize changes in the depositional environment during the ontogenetic development of the Lake Temje. The analysis of fossil chironomid remains and statistical treatment of chironomid data by the application of a newly developed regional Russian transfer functions provided inferences of mean July air temperatures (T July ) and water depths (WD). Reconstructed WDs show minor changes throughout the core and range between 80 and 120 cm. All the fluctuations in reconstructed water depth lie within the mean error of prediction of the inference model (RMSEP ¼ 0.35) so it is not possible to draw conclusions from the reconstructions. A qualitative and quantitative reconstruction of Holocene climate in central Yakutia recognized three stages of palaeoenvironmental changes. The early Holocene between 10 and 8 ka BP was characterized by colderthan-today and moist summer conditions. Cryotextures in the lake sediments document full freezing of the lake water during the winter time. A general warming trend started around 8.0 ka BP in concert with enhanced biological productivity. Reconstructed mean T July were equal or up to 1.5 C higher than today between 6.0 ka and 5.0 ka BP. During the entire late Holocene after 4.8 ka BP, reconstructed mean T July remained below modern value. Limnological conditions did not change significantly. The inference of a mid-Holocene climate optimum supports scenarios of Holocene climatic changes in the subpolar part of eastern Siberia and indicates climate teleconnections to the North Atlantic realm.
