Chronic Exposure to Environmentally Relevant Concentrations of Microcystin-Leucine Arginine Causes Lung Barrier Damage through PP2A Activity Inhibition and Claudin1 Ubiquitination

Abstract: Microcystin-leucine arginine (MC-LR), ubiquitous in water and food, is a threat to public health. In the present study, after C57BL/6J mice were fed with environmental concentrations of MC-LR (0, 1, 30, 60, 90, and 120 μg/L) for 6, 9, and 12 months, it was found that MC-LR could enter into mouse lung tissues and cause microstructural damage, as shown by western blotting and HE staining. Electron microscopy examination showed that MC-LR could damage the lung barrier by disruption of the tight junctions, which w… Show more

“…The contamination of MCs and As is becoming increasingly serious to the development of the social economy [ 38 , 39 , 40 , 41 ]. The median exposure level of MCs and As were 0.16 μg/L and 1.12 μg/L, respectively, indicating that subjects were exposed to MCs and As.…”

“…It has been found more than 279 microcystin variants, and microcystin‐LR (MC‐LR) is one of the most widely distributed and most toxic congeners 17,18 . MC‐LR has adverse effects on liver, kidney, gonad and other organs after entering the body 19,20 . Importantly, MC‐LR has strong hepatotoxicity 21 .…”

“…3 Microcystin-LR (MC-LR) is the most ubiquitous and toxic metabolites released by cyanobacteria during eutrophication. 4 Even low concentrations, MC-LR may cause damage to liver and kidney and render tumor promotion by inhibiting protein phosphatases. 5 Considering its ubiquity and high toxicity, the World Health Organization (WHO) has set the maximum allowable concentrations for MC-LR to be 1 nM in drinking water.…”

“…Algal blooms have been increasing globally due to the climate changes, water eutrophication, and rinsing agricultural activities. , As the most pressing global environment problems, cyanobacteria blooms cause a tremendous threat to food safety, water resources, and human health . Even low concentrations, MC-LR may cause damage to liver and kidney and render tumor promotion by inhibiting protein phosphatases .…”
