Chromosome Dynamics during Mitosis

Abstract: The primary goal of mitosis is to partition duplicated chromosomes into daughter cells. Eukaryotic chromosomes are equipped with two distinct classes of intrinsic machineries, cohesin and condensins, that ensure their faithful segregation during mitosis. Cohesin holds sister chromatids together immediately after their synthesis during S phase until the establishment of bipolar attachments to the mitotic spindle in metaphase. Condensins, on the other hand, attempt to "resolve" sister chromatids by counteracting… Show more

“…Shugoshin‐1 (Sgo1) protects cohesin proteins and centrosome integrity (Salic, Waters, & Mitchison, 2004; Schöckel, Möckel, Mayer, Boos, & Stemmann, 2011). Cohesins keep sister chromatids from prematurely separating during mitosis, thus ensuring mitotic fidelity (Hirano, 2015). The Sgo1 haploinsufficient (∓) mouse is a model of cohesinopathy and chromosome instability (CIN) (Rao et al., 2016; Wang et al., 2008; Yamada et al., 2012, 2015, 2016).…”

“…Meiosis-specific cohesin components, including SMC1β, two α-kleisins (REC8 and RAD21L) and a stromal antigen protein (STAG3), are important for these additional requirements of cohesins during meiosis (Bannister et al, 2004;Fukuda et al, 2014;Herrán et al, 2011;Hopkins et al, 2014;Llano et al, 2014;Revenkova et al, 2004;Winters et al, 2014;Xu et al, 2005). Mutation of meiosisspecific cohesin components in female mice results in an increased frequency of oocyte aneuploidy and premature ovarian failure The two condensin complexes (I and II) are composed of the SMC2 and SMC4 heterodimers, but their kleisin subunit and pair of HEAT repeat elements are unique (Hirano, 2015). Condensins localize to the longitudinal axes of bivalents following meiotic resumption in mouse oocytes, and both complexes are required for chromosome compaction before meiosis I (Houlard et al, 2015;Lee et al, 2011).…”

“…In parallel to this, the cohesin complex is deposited along the chromosomes to encircle and hold the sister chromatids together (Tanaka et al 2001;Nasmyth 2011). Most of the DNA catenanes are removed by topoisomerase IIα (TopIIα); either during S phase or during early mitosis when DNA condensation occurs (Hirano 2015). In the prophase of mitosis, most of the cohesin located on chromosome arms (but not at centromeres) is released in a condensation-dependent manner (Hirano 2015).…”
