Chromatography of RNA-DNA Complexes on Hydroxyapatite. A Method for the Separation of the Complementary Strands in T2 DNA

Abstract: The fractionation of nucleic acids by chromatography on hydroxyapatite has been studied. The products, formed by annealing T2 DNA with complementary RNA, were separated in a linear gradient of phosphate buffer into three fractions containing, respectively, in their order of elution, single-stranded DNA, RNA complexed mainly with single-stranded DNA, and renatured DNA complexed with smaller amounts of RNA. By using annealing conditions which favour hybrid formation the complementary strand of DNA can be separat… Show more

“…(Log Me, is the appropriate quantity to use when comparing elution molarities, Martinson, 1973b. ) The chromatographic behaviors of some types of DNA-RNA hybrid molecules on hydroxylapatite have been reported (see in particular Becker et al, 1970, andSiebke andEkren, 1970). However the hybrids of Becker et al (1970) contained only 25 % or less RNA and the studies of Siebke and Ekren (1970) involved T2 phage DNA which chromatograms atypically on hydroxylapatite (Oishi, 1971).…”
