The authors wish to congratulate Professor Walther Lamprecht, Hannover, on the occasion of his sixtieth birthday Summary:We describe an on-line multi-cplumn high performance liquid Chromatographie method for the selective clean-up and analysis of major and minor ribonucleosides in physiological fluids. Quantitative data obtained for the determination of some methylated ribonucleosides in human urines are compared with those obtained with the traditional off-line method.The on-line technique developed in our laboratory is distinguished from the off-line method by the following features:(1) Sample clean-up and analysis of the target-compounds can easily be automatized, (2) Total time of analysis, for example of urinary ribonucleosides, is decreased to 35 minutes, (3) Laborious and error-prone evaporation and redissolution Steps are avoided, (4) Reliability of the overall analytical System can be controlled with ease, (5) Small sample-volumes can be applied directly, (6) Sensitive samples can be processed very rapidly under mild conditions, (7) Results obtained with the -line and off-line-techniques compare well. Der NAD(P)-Coenzymmetabolit 1,6-Dihydr0-6-oxo-l -(P-~-ribofuranosyl)-3-pyridincarbonsaureamid (9), der im menschlichen Harn ausgeschieden wird, und die entsprechende 3-Carbonsaure 8 wurden durch Kondensation von silyliertem 6-0x0-3-pyridincarbonsaureamid 4 sowie dem Silylderivat 2 der entsprechenden 3-Carbonsaure mit 1,2,3,5-Tetra-O-acetyl-~-~-ribofuranose in Acetonitril bei Gegenwart von Zinntetrachlorid synthetisiert. Die Ribosidierungsposition in 8 und 9 wurde durch H-und '3C-NMR-Spektroskopie bestirnmt. Synthesis of the Coenzyme Metabolite l,Q-Dihydro-6-oxo-l-(~-~-ribofuranosyl)- 3-pyridinecarboxamideThe NAD(P) coenzyme metabolite 1,6-dihydro-6-oxo-l-(~-~-ribofuranosyl)-3-pyridinecarboxamide (9) which is excreted in human urine and its 3-carboxylic acid 8 were synthesized by condensation of silylated 6-0x0-3-pyridinecarboxamide 4 and the corresponding silylated 3-carboxylic acid 2 with 1,2,3,5-tetra-O-acetyl-~-~-ribofuranose in acetonitrile in the presence of tin tetrachloride. The ribosidation site in 8 and 9 was assigned by 'H and I3C NMR spectroscopy. Als weiteres naturlich vorkomrnendes Pyridinnucleosid ist bisher nur das toxische Clitidin (12) aus dem japanischen Pilz CIirocybe ugromelulga isoliert und synthetisiert wordens-'). Bei Verzehr des Pilzes verursacht Clitidin schmerzhafte erythromelalgieahnliche Symptome an Handen und FiiBen5 -7 ) . Strukturell ist 12 als 1,4-Dihydro-4-imino-l-(~-~-ribofuranosyl)-3-pyridincarbonsaure dem harngangigen Coenzymmetaboliten 9 eng verwandt, so dal3 die chemische Synthese von 9 auch aus pharmakologischer Sicht interessant ist. Analysis of the modified nucleosides is particularly important in the medical area because of a possibility of cancerogenic processes studies. The aim of this work was to study the selectivity tuning of modified nucleosides through the investigations of interactions analyte (modified nucleoside) <==> stationary phase <==> mobile phase. A series of homemade stationary phases with different surface properties has been utilized. All of them contain various interaction sites such as: cholesterol (SG-CHOL); n-acylamide (SG-CHOL, SG-AP); aminopropyl (SG-CHOL, SG-AP, SG-NH2, SG-MIX); cyanopropyl, phenyl, octyl (SG-MIX), octadecyl (SG-MIX, SG-C18) and silanols localized on the silica gel surface of all packings. The attempt to predict the main interactions responsible for the retention between nucleosides and stationary phase ligands was done on the basis of the elemental analysis, and proportional part of an individual ligand bonded to silica surface results. In order to study the influence of different packing types on the analyzed nucleosides retention, the relationship between pH of the mobile phase buffer and the selectivity of a stationary phase was investigated.
