Aerobic microorganisms were isolated and identified from 9 of 10 malignant tissues aseptically obtained from surgical patients. The organisms isolated are species commonly associated with the flora of the human body. When these cancer-associated organisms were grown in Trypticase soy broth (BBL Microbiology Systems), a protein substance was isolated from the culture filtrates by acetone precipitation. The acetone precipitates of 12 of 14 organisms tested were positive when assayed by radioimmunoassay for the beta subunit of human chorionic gonadotropin (hCG). All but one of the bacterial isolates from the malignancies were capable of producing the hCG-like substance, but in varying quantities. Control organisms (not isolated from a malignancy) and uninoculated Trypticase soy broth were either completely negative in the radioimmunoassay for beta hCG or had levels of beta hCG near the limit of the sensitivity of the method. These results suggest the possibility that bacteria-tumor relationships do exist and are in agreement with the findings of other workers. The presence of choriogonadotropinand a-subunit-like materials in two species of bacteria identified as Staphylococcus simulans and Streptococcus faecalis have been demonstrated by the indirect fluorescein-labeled and the indirect peroxidase-labeled immunocytochemical techniques, utilizing antiserum for human choriogonadotropin, for its a and ,8-subunits and the fl-subunit COOHterminal peptide. The bacteria were originally isolated from the urine of two patients with advanced forms of cancer. Chromatography done on the watersoluble extract of acetone powder preparations of the bacterial cultures revealed the presence of a material similar to the complete trophoblastic hormone and to its fl-subunit in the culture media of S. simulans, and to the fl-subunit in the media of S. faecalis. No free a-subunit was detectable. Furthermore, the choriogonadotropin-like factor demonstrated biological activity in in vivo assay systems. From the present results, it can be concluded that some species of "cancerassociated" bacteria can synthesize a human trophoblastic hormone-like glycoprotein with physicochemical properties similar to those of the human trophoblastic hormone that is biologically active and that is either released complete or as one of its subunits in the culture media. 487 on July 31, 2020 by guest http://iai.asm.org/ Downloaded from

To determine whether the in vivo production of antibodies against choriogonadotropin (CG) could modify the relationship between host and malignant growth, the effects of preimmunization with a conjugate of the β‐subunit of CG and tetanus toxoid (CGβ‐tt) on the development and growth of lung metastasis following I.V. injection of R 3230 rat mammary adenocarcinoma cells into Fischer 344 female rats have been investigated. This neoplasm has been previously demonstrated to synthesize CG‐like material. A test group of 70 animals and two matched control groups of 15 animals each were used. One of the control groups was untreated and the other received tetanus toxoid only. All the control animals had multiple lung foci of neoplastic cells ten days after seeding, as has been previously observed, and CG antibodies were undetectable. In contrast, a significant titer of anti‐CG was found in all preimmunized animals. At the same time, the pretreated animals rarely had a few small neoplastic nodes in the entire lung sections at the standard 8–10 days post‐seeding. The protective effects of preimmunization with CGβ‐tt were further demonstrated by the animals living 20 days post‐seeding, the absence of lung pathology in the ones killed thereafter, and by six animals that were left alive and have not shown any deterioration for more than six months after the administration of the cell suspension.
