Chiral separation and quantitation of cetirizine and hydroxyzine by maltodextrin‐mediated CE in human plasma: Effect of zwitterionic property of cetirizine on enantioseparation

Abstract: In the present study, a very simple CE method for chiral separation and quantitation of zwitterionic cetirizine (CTZ), as the main metabolite of hydroxyzine (HZ), and HZ has been developed. In addition, the effect of zwitterionic property of CTZ on enantioseparation was investigated. Maltodextrin, a linear polysaccharide, as a chiral selector was used and several parameters affecting the separation such as pH of BGE, concentration of chiral selector and applied voltage were studied. The best BGE conditions for… Show more

“…Electroosmotic flow (EOF), Joule heating and the adsorption of analyte on the inner surface of capillary column, are the phenomena which were influenced by buffer concentration [35]. The 0.6% (w/v) CM-β-CD and 30% (v/v) methanol at pH 2.5 BGE were applied for the optimization of phosphoric acid-TEA buffer (30-100 mM) concentration.…”

“…Temperature variations affect enantioseparation with different mechanisms, for example, buffer viscosity, Joule heating, migration times and complex formation equilibria between enantiomers and chiral selector [35,36]. Several capillary column temperatures (15, 20 and 25°C) were checked.…”

“…Thus, this sample treatment continues being one of the most employed. Table shows the use of LLE for the enantiomeric determination of drugs in biological samples as plasma or microsomal fraction of liver homogenates , and of fungicides in environmental samples as slurries . After the sample treatment, LODs from 7 × 10 −8 to 5 × 10 −6 M were reported when UV detection was used.…”
