Chikungunya Virus Infection Results in Higher and Persistent Viral Replication in Aged Rhesus Macaques Due to Defects in Anti-Viral Immunity

Abstract: Chikungunya virus (CHIKV) is a re-emerging mosquito-borne Alphavirus that causes a clinical disease involving fever, myalgia, nausea and rash. The distinguishing feature of CHIKV infection is the severe debilitating poly-arthralgia that may persist for several months after viral clearance. Since its re-emergence in 2004, CHIKV has spread from the Indian Ocean region to new locations including metropolitan Europe, Japan, and even the United States. The risk of importing CHIKV to new areas of the world is increa… Show more

“…These longitudinal analyses suggest that functional T and/or B cell responses protect against chronic musculoskeletal disease. Our results are consistent with recent findings in rhesus macaques in which persistence of CHIKV in the spleen correlated with defects in adaptive immune responses (54) and with findings in humans in which the rapid appearance of neutralizing IgG3 antibodies correlated with viral clearance and protection from chronic CHIKV disease (55).…”

“…(i) Rhesus macaques. Viral RNA load in the tissues at necropsy of the rhesus macaques was quantified using real-time reverse transcription-PCR (RT-PCR) (38). Primers and probes included the following: CHIKV-9482F, 5=-GGAACGAGCAGCAACCTT TG-3=; CHIKV-9931R, 5=-ATGGTAAGAGTCTCAGACAGTTGCA-3=; and the TaqMan probe CHIKV-9870F, 5=-GGAATAAGGGCTTGT-3=.…”

“…In cynomolgus macaques, CHIKV persists in joints, muscles, lymphoid organs, and liver for at least 3 months (37), providing a possible explanation for the long-lasting CHIKV symptoms observed in humans. While adult rhesus macaques effectively control CHIKV infection, aged animals developed persistent infection in the spleen (38).…”

“…8, 72, and Figure 3). Persistent CHIKV infection also has been observed for weeks to months in experimentally infected mice (137,147) and NHPs (105,163), with vRNA detected in joint tissue (150), muscle, and spleen (105). In addition, persistent IgM levels have been detected in patients and animals experiencing chronic arthritis (8,105,126,153,164,165).…”
