Chikungunya Virus Envelope-Specific Human Monoclonal Antibodies with Broad Neutralization Potency

Abstract: Chikungunya virus (CHIKV) is an alphavirus responsible for numerous epidemics in Africa and Asia. Infection by CHIKV is often characterized by long-lasting, incapacitating arthritis, and some fatal cases have been described among elderly and newborns. Currently, there is no available vaccine or specific treatment against CHIKV. Blood B cells from a donor with history of CHIKV infection were activated, immortalized, amplified, and cloned. Two human mAbs against CHIKV, 5F10 and 8B10, were identified, sequenced, … Show more

“…However, CHIKV with a V216E or R82G mutation on E2 were also able to propagate in the presence of antibody 5F10 (16). Similarly, CHIKV with mutations T12I or R82G on domain A of E2 or T101M on domain II of E1 (16) were able to propagate in the presence of antibody 8B10.…”

“…The CHIKV-neutralizing antibodies 8B10 and 5F10 had been previously isolated from CD-40-activated B cells of infected patients (15) and have been shown to protect mice from CHIKV infection (16). However, CHIKV with a V216E or R82G mutation on E2 were also able to propagate in the presence of antibody 5F10 (16).…”

“…An IgG response can be detected approximately 7-10 days after onset of illness, often after viremia has been cleared (37,(154)(155)(156). Many mouse and human IgM and IgG antibodies broadly and potently neutralize CHIKV (157)(158)(159)(160)(161)(162). The most potently neutralizing antibodies target domains A and B of the E2 glycoprotein, with those targeting domain B often displaying broad neutralization against multiple strains of CHIKV and other related alphaviruses (157,(159)(160)(161).…”

“…Many monoclonal antibodies (mAbs) that neutralize CHIKV infection recognize epitopes on the solvent-accessible surface of the E2 protein (28)(29)(30)(31)(32). The only previous structural studies of CHIKVantibody complexes were of four neutralizing murine mAbs.…”
