Chemically-Induced Colitis Models in Animal

Abstract: Ulcerative colitis is a chronic inflammation that can affect the distal part of the colon, submucosa and rectal mucosa, and can affect the entire colon even to the terminal ileum. There are several factors that can cause this disease, such as genetics, environment, intestinal microbiota and the presence of enteric infectious agents. Chemical induction in experimental animals for research on gastrointestinal inflammation has been frequently used due to the similarity of the anatomical and physiological structur… Show more

“…More importantly, compared to the DSS group, colitis mice in the 10 mg/kg LP + DSS group exhibited a significant increase in body weight (P < 0.01). In terms of clinical symptoms, DSS-induced colitis has typical symptoms similar to those of human colitis, such as weight loss, diarrhea, and blood in the stools [18]. As shown in Figure 1b, DAI in the DSS group was significantly higher than that in the normal group (P < 0.01).…”

“…The UC model induced by dextran sulfate sodium (DSS) is one of the most widely used UC models and has the advantages of a simple method and high success rate. 18 C57BL/6 mice are severely sensitive to DSS, and at the lesion site, the clinical manifestations and pathological manifestations in these mice are very similar to those of human UC. 19 In this study, DSS induction was used to establish a classic UC mouse model, and by observing the effect of different doses of LP on clinical symptoms in this model, we were able to simultaneously determine the antioxidant enzyme activity; inflammatory cytokine secretion; and TLR4, TRIF, p-NF-κB p65, ZO-1, claudin-1, and occludin protein expression in colon tissues.…”

“…We selected DCA, because it has been reported to inhibit NLRP3 activation (Berg 2020 ; Forteza et al 2023 ). Next, we established chemically induced colitis model using oxazolone as previously reported in BALB/c mice (Siregar and Widyawati 2021 ) followed by testing the effect of DCA on alleviating oxazolone-induced colitis. Colitis was induced in BALB/c mice using topical 3% oxazolone solution (day 0) followed by enema of 1% oxazolone solution dissolved in ethanol (day 5).…”

“…Acute ulcerative colitis was induced in BALB/c mouse strain that are susceptible to Th2 immune activation as previously described (Siregar and Widyawati 2021 ; Wirtz et al 2007 ). That was initiated by pre-sensitization of mice through percutaneous application of 150 μL 3% oxazolone (4-ethoxymethylene-2-phenyl-2-oxazolin-5-one, CAT# 862207, Sigma-Aldrich, St Louis, MO, USA) solution at a dilution (4:1 in acetone and organic extra virgin olive oil) on the animal’s 2 × 2 cm field shaved abdomen.…”
