Summary Strigolactones (SLs) are plant hormones that regulate diverse developmental processes and environmental responses. They are also known to be root‐derived chemical signals that regulate symbiotic and parasitic interactions with arbuscular mycorrhizal fungi and root parasitic plants, respectively. Since the discovery of the hormonal function of SLs in 2008, there has been much progress in the SL research field. In particular, a number of breakthroughs have been achieved in our understanding of SL biosynthesis, transport and perception. The discovery of the hormonal function of SL was quite valuable not only as the identification of a new class of plant hormones, but also as the discovery of the long‐sought‐after SL biosynthetic and response mutants. These mutants in several plant species provided us the genetic resources to address fundamental questions regarding SL biosynthesis and perception. Such mutants were further characterized later, and biochemical analyses of these genetically identified factors have uncovered the outline of SL biosynthesis and perception so far. Moreover, new genes involved in SL transport have been discovered through reverse genetic analyses. In this review, we summarize recent advances in SL research with a focus on biosynthesis, transport and perception.

Strigolactones are plant hormones and rhizosphere signalling molecules with key roles in plant development, mycorrhizal fungal symbioses, and plant parasitism. Currently, sensitive, specific, and high-throughput methods of detecting strigolactones are limited. Here, we developed genetically encoded fluorescent strigolactone biosensors based on the strigolactone receptors DAD2 from Petunia hybrida, and HTL7 from Striga hermonthica via domain insertion of circularly permuted GFP. The DAD2 biosensor exhibited loss of cpGFP fluorescence in vitro upon treatment with the strigolactones 5deoxystrigol and orobanchol, or the strigolactone analogue GR24. The biosensor likewise responded to strigolactones in an in vivo protoplast system, and retained strigolactone hydrolysis activity. The ShHTL7 biosensor exhibited loss of cpGFP fluorescence upon GR24 treatment in vitro, and responded to a specific inhibitor of ShHTL7 but not DAD2, indicating that the biosensors retained the specificity of their parent receptors. These biosensors have applications in high-throughput screening, and may also have utility for studying strigolactone biology.

Today, the use of artificial pesticides is questionable and the adaptation to global warming is a necessity. The promotion of favorable natural interactions in the rhizosphere offers interesting perspectives for changing the type of agriculture. Strigolactones (SLs), the latest class of phytohormones to be discovered, are also chemical mediators in the rhizosphere. We present in this review the diversity of natural SLs, their analogs, mimics, and probes essential for the biological studies of this class of compounds. Their biosynthesis and access by organic synthesis are highlighted especially concerning noncanonical SLs, the more recently discovered natural SLs. Organic synthesis of analogs, stable isotope‐labeled standards, mimics, and probes are also reviewed here. In the last part, the knowledge about the SL perception is described as well as the different inhibitors of SL receptors that have been developed.
