Chemical Functionalization of Bioceramics To Enhance Endothelial Cells Adhesion for Tissue Engineering

Abstract: To control the selective adhesion of human endothelial cells and human serum proteins to bioceramics of different compositions, a multifunctional ligand containing a cyclic arginine-glycine-aspartate (RGD) peptide, a tetraethylene glycol spacer, and a gallate moiety was designed, synthesized, and characterized. The binding of this ligand to alumina-based, hydroxyapatite-based, and calcium phosphate-based bioceramics was demonstrated. The conjugation of this ligand to the bioceramics induced a decrease in the n… Show more

“…The preparation of cRGDfK (8) was performed by solid phase peptide synthesis as previously described. 33 The introduction of an alkynyl moiety was carried out by coupling with activated ester of pentynoic acid to afford intermediate 9 in 61% yield (Scheme 3). The azide-alkyne Huisgen's cycloaddition reaction was designed to occur at the interface of azide-PEG coating of the NPs suspended in H 2 O. Ligand 9, dissolved in DMF, was added to a suspension of cNPs and the mixture was ultrasonicated for 12 h in the presence of copper sulfate and sodium ascorbate.…”

“…It can mixed dissolve with water, ethanol, hydrocarbon and oils. TTEG's production and used as a solvent, plasticizer, lubricant, softening agent for paper tissue, a ceramic paste [3] printing ink binder and a liquid desiccant for natural gas. Moreover, tetraethylene glycol to the eye and the upper respiratory tract has slightly stimulation [4].…”

“…This observation was in accordance with previous results obtained for other functionalizing ligands. [15] In conclusion, surface functionalization of HA and TCP biomaterials provides an efficient tool to promote the adhesion of bone precursor cells to the scaffolds using the copper free click reaction. The biocompatibility of this chemical transformation opens the way to further development of functionalized bioceramics as cell-engineered bone implant materials.…”
