Chemical fingerprinting and quantitative monitoring of the doping drugs bambuterol and terbutaline in human urine samples using ATR-FTIR coupled with a PLSR chemometric tool

Abstract: ATR-FTIR spectroscopy coupled with chemometric tools could be a suitable alternative to the traditional techniques for quantification of the performance enhancing drugs such as bambuterol and terbutaline in urine samples in and out of competition.

“…SEIRA bands are based on the chemical functional groups of analytes as each function group has its unique absorption band within the active mid-IR range (4600–400 cm −1 ); consequently, the SEIRA spectrum is considered a fingerprint for molecules [ 31 ]. These fingerprints can be used for the identification and differentiation of molecules [ 32 ].…”

“…Furthermore, repeating the sampling process for confirmation purposes is also easier in case of urine [87,88]. The potential application of ATR-FTIR in the detection and quantification of doping agents in urine samples has been investigated [89,90]. Terbutaline and its precursor bambuterol are tow drugs used in asthma treatment in more than 25 countries [91].…”

“…In 2020, Algethami et al explored the potential use of ATR-FTIR in combination with chemometrics in this type of analyses. It has been demonstrated that this technique can detect and quantify these two drugs in volunteers' urine with a high level of accuracy [89]. Moreover, MT-45 is a synthetic opioid that has been developed in the years 1970s by a Japanese pharmaceutical company to be used as an alternative painkiller for morphine.…”

“…[24] Furthermore, this technique has application, in combination with multivariate modelling techniques, for the detection of banned substances in urine. [25] The Amide II band, due to CÀ N stretching and NH 2 bending, correlates well to albuminuria levels at high concentrations. [26] However, the prediction of proteins from the direct measurement of urine or dried urine is strongly limited by their low concentration and the presence of major interference such as urea and creatinine.…”

“…We have recently demonstrated that mid‐IR transflection spectroscopy can also quantify 8‐isoprostane, a marker of ROS, in urine [24] . Furthermore, this technique has application, in combination with multivariate modelling techniques, for the detection of banned substances in urine [25] . The Amide II band, due to C−N stretching and NH 2 bending, correlates well to albuminuria levels at high concentrations [26] .…”
