This work aims to evaluate the synergetic antimicrobial effects of cold nitrogen plasma (CNP) and lemongrass oil against L. monocytogenes on pork loin. Their possible effects on the physicochemical properties of pork were investigated as well. Lemongrass oil displayed anticipant antibacterial activities against L. monocytogenes at high dose, which could damage the bacterial membrane, leading to the losses of intracellular constituents, including DNA, protein and ATP. Compared with control groups, the number of L. monocytogenes on pork samples was reduced by 2.80 log 10 steps after synergy treatment of CNP (500 W, 120 s) and lemongrass oil (5 mg/mL, 30 min). With the assistance of CNP, the satisfactory antibacterial effect for L. monocytogenes on pork was achieved after treatment for low concentration of lemongrass oil. In addition, the potential impacts on sensory attributes of pork were also minimized via combined treatment. Practical applicationsUnder low concentration, lemongrass oil achieved the desired antibacterial effect for L. monocytogenes on pork loin with the assistance of CNP. The potential impacts on sensory attributes of pork loin were minimized via synergetic treatment as well. The synergetic treatment provides a new way for application of plant essential oil in food preservation. Six lignans including two new lignans were obtained as the principal components of the Forsythia koreana flowers via silica gel (SiO ), octadecyl SiO (ODS) as well as Sephadex LH-20 column chromatography. In addition to two new lignans, named koreanaside A ((7R,8S,7'R,8'S)-7,7'-diepoxy-5'-hydroxy-3,3'-dimethoxylignan 4-O-β-d-glucopyranoside) and koreanaside B ((7R,8S,7'S,8'R)-7,9'-epoxy-9,5',7'-trihydroxy-3,3'-dimethoxylignan 4-O-β-d-glucopyranoside), four known lignans were identified to be (+)-phylligenin, (-)-epipinoresinol, pinoresinol, and tinosposide A. The structures and absolute configurations of koreanasides A and B were established by means of analysis of spectroscopic data (NMR, IR, FAB-MS, and CD), whereas the structures of known lignans were identified by comparison their NMR and MS values with those in the reported literature. Their chemical structures including configuration were established by means of analysis of spectroscopic data (NMR, IR, FAB-MS, and CD) but also comparison of their NMR and MS values with those in the reported literature. This is the first article for isolation of six lignans of F. koreana flowers. Koreanasides A and B showed high radical scavenging activity with oxygen radical absorbance capacity (ORAC) values of 0.97 ± 0.01 and 1.02 ± 0.01, respectively. Koreanaside A also prohibited expressing VCAM-1 in MOVAS cells with 80.5% at 25 mg/mL.

Prompt elimination of pathogens including bacteria and dead cells prevents the expansion of secondary and prolonged inflammations and tissue damage. Keigairengyoto (KRT) is a traditional Japanese medicine prescribed for dermatoses such as purulent inflammations. Our aim is to clarify the actions of KRT in bacterial clearance and to examine the cell-kinetic profiles of phagocytes. In a mouse cutaneous infection model using living Staphylococcus aureus, KRT drastically reduced the number of bacteria in the infection sites. To evaluate the bacterial clearance, pseudo-infection was induced in mouse ears by intradermal injection of FITC-conjugated dead S. aureus. Biochemical and histological examinations revealed that KRT promoted bacterial clearance at 6 and 24 h post-injection. The numbers and phagocytic activities of neutrophils and macrophages in the ears were evaluated histologically using anti-Ly6G and F4/80 antibodies. KRT reduced bacterial deposition and increased the accumulation of F4/80 + resident macrophages around the lesion site. FACS analysis was performed on single cell suspensions dispersed enzymatically from skin lesions, followed by an investigation of CD11b + Ly6G + (neutrophils) and CD11b + Ly6G -(monocytes/macrophages) cells. KRT increased the mean fluorescent intensity of FITC in CD11b Ly6G-cells and the number of FITCpositive CD11b + Ly6G+ cells, while KRT did not change the numbers of these cells. To investigate the active constituents of KRT, phagocytosis assay using macrophages was performed, resulting in that some flavonoid glucuronides of KRT derivatives augmented phagocytosis. Collectively, KRT promoted bacterial clearance by enhancing the phagocytic capability of neutrophils and macrophages. KRT may exert unique properties in preventive and therapeutic strategies for skin infectious inflammation.
