Chelerythrine Attenuates Renal Ischemia/ Reperfusion-induced Myocardial Injury by Activating CSE/H2S via PKC/NF-κB Pathway in Diabetic Rats

Abstract: Background/Aims: Chelerythrine (CHE), a benzophenanthridine alkaloid, is a potent, selective, and cell-permeable protein kinase C (PKC) inhibitor. The purpose of the present study was to evaluate the effect of CHE on myocardial recovery after renal ischemia/reperfusion (I/R)-induced myocardial injury (RI/RMI) in a streptozocin (STZ)-induced diabetic rat model. Methods: Diabetes mellitus (DM) rats preconditioned with CHE and D, L-propargylglycine (PAG) were subjected to renal I/R. The extent of cardiac morpholo… Show more

“…It was found in our study that the activity of SOD and GSH level were decreased while the MDA level was increased in the renal tissue in diabetic RI/RI, demonstrating an oxidative injury. During the process of I/R injury, inflammatory reactions are activated, resulting in the formation of inflammatory cytokines, such as IL-6 and IL-10 [9, 27, 43, 44]. We also found that the expressions of IL-10 and IL-6 in the renal tissue were elevated in this diabetic RI/RI model.…”

“…injection of STZ dissolved in 0.1 M citrate buffer (PH 4.5, 50 mg/kg body weight [bw]). Rats with serum glucose levels ≥16.7 mmol/L in three consecutive measurements (three consecutive days) were considered to be a successful diabetic rat model [27]. …”

“…Myocardial ischemia-reperfusion (I/R) injury (MI/RI) as a common pathophysiological entity of cardiovascular diseases is one of the leading causes of death worldwide [1,2]. Lipid peroxidation, reactive oxygen species (ROS) production, apoptosis, and DNA/RNA injury are reported to be associated with MI/RI [3].…”

“…As described previously [15, 29, 30], the serum activities of LDH and CK were measured using commercial assay kits according to manufacturer’s instructions. The activity of LDH was expressed as U of LDH/L and the activity of CK was expressed as U of CK/L.…”

“…Autophagy is an important pathological and physiological phenomena in which external stimuli or starvation causes components or structures of eukaryotic cells to be enveloped in double membrane vesicles (autophagic vesicles) that subsequently fuse with the lysosomes and are eventually hydrolyzed by the lysosomes [15]. Autophagy, as one of the lysosomal degradation pathways, plays an important role in the half-life of protein degradation and cell renewal [16].…”
