Characterization of Zebrafish Pax1b and Pax9 in Fin Bud Development

Abstract: Both Pax1 and Pax9 belong to the important paired box gene family (PAX), which mainly participates in animal development and sclerotome differentiation. To date, the precise molecular mechanism and related signaling pathway of Pax1 remain unclear. In our study, microinjection of morpholino- (MO-) modified antisense oligonucleotides against pax1b induced pectoral fin bud defects. Furthermore, we demonstrate that the phenotypes caused by the knockdown of Pax1b in zebrafish could not be phenocopied by pax9 MO and… Show more

“…In addition to mice, zebrafish are also suitable models for cancer research. In the literature, zebrafish have been used successfully to elucidate the role of pax9 in the development of blood cells [ 41 , 72 ], oro-esophageal squamous epithelium [ 30 ], fin [ 73 ], tooth [ 74 ], and palate [ 75 ].…”

“…Its conserved roles can be demonstrated by similar observations in different species. For example, Pax1 is essential for fin bud development in zebrafish, wing bud development in chick and limb bud development in mouse embryos ( Timmons et al, 1994 ; Hofmann et al, 1998 ; Chen et al, 2014 ). Interestingly, as a transcription factor in the same subfamily with similar protein domains, PAX9 was often investigated in parallel with PAX1.…”

“…Embryonic chick wings show morphological defects in the shoulder girdle when Pax1 expression is reduced by ectopic application of signaling molecules (BMP2 and BMP4) ( Hofmann et al, 1998 ), which parallel the defects seen in Pax1 mutant mice ( Timmons et al, 1994 ; Dietrich and Gruss, 1995 ). In zebrafish, microinjection of morpholino- (MO-) modified antisense oligonucleotides against pax1b induced pectoral fin bud defects which could not be phenocopied by pax9 MO and could not be rescued by either pax1a or pax9 overexpression, whereas could be partially rescued by mouse Pax1 mRNA injection ( Liu et al, 2013 ; Chen et al, 2014 ). Loss-of-function of pax1b in zebrafish affects the expression of col2a1 , uncx4.1 , noggin3 and aggrecan , suggesting its roles in chondrocytes differentiation ( Chen et al, 2014 ).…”

“…Among these are twist , pax1 , pax9 , nkx3.1 and nkx3.2 (Akazawa et al 2000 ; Balling et al 1996 ; Barnes and Firulli 2009 ; Gitelman 1997 ; Herbrand et al 2002 ; Peters et al 1999 ; Provot et al 2006 ; Rainbow et al 2014 ; Tribioli and Lufkin 1999 ). Expression of these markers has been shown to be conserved in the zebrafish sclerotome (Germanguz et al 2007 ; Nornes et al 1996 ; Liu et al 2013 ; Ma et al 2018 ), but only very rarely has their involvement in specification and development of this somitic compartment been investigated using this model organism (Yang et al 2011 ; Ma et al 2018 ; Chen et al 2014 ).…”
