Characterization of the PEG layer of sterically stabilized liposomes: a SAXS study

Abstract: Synchrotron small-angle X-ray scattering analysis of the bilayer structure of a pharmacologically relevant sterically stabilized liposome system is presented. Describing the electron density profile of the bilayer with the superposition of Gaussian functions, the contribution of the poly(ethylene glycol) (PEG) layers to the total electron density was identified. The changes in the thickness of the PEG layer as well as the distribution of the PEG chains among the outer and inner leaflets of the bilayers were fo… Show more

“…Based on small angle X-ray scattering study, the thickness of the PEG 2000 layer on liposomes was estimated to be ~5 nm when the PEG 2000 fraction is 10 mol%. 34 The self-assembly of 2 gave a broad peak with d-spacing of 5.5 nm as measured by the present powder XRD analysis. The difference in thickness of each PEG moiety was attributable to difference in density of PEGylation and morphology, which may cause deformation of random coils of PEG.…”

“…Thus, they developed a moderated PEGylated (size 2 K) multicomponent liposome that hindered protein adsorption but still presented high cellular uptake in cancer cells [74]. Therefore, all of these studies were relevant for the development of efficient biohybrid liposomal pharmaceutical forms, contributing to the elucidation of their performance in the bloodstream [75,76].…”

“…Numerous attempts have been made to graft PEG in the bilayer surface for investigating the structure and nature of the bilayer membrane. However, most of the studies so far are limited to the nanometer scale [2,3] because of the instability of liposome bilayer in macroscale.…”
