Characterization of Pleurotus djamor neohaplonts recovered by production of protoplasts and chemical dedikaryotization

Abstract: Production of hybrid strains is accomplished by mating monosporic isolates or neohaplonts, obtained either by chemical dedikaryotization or by production of protoplast. However, differences in growth rate among recovered neohaplonts have been reported. The presence of phenotypic and genetic changes among the neohaplonts recovered either by chemical dedikaryotization or by production of protoplast, was evaluated by measuring growth and morphology, and by molecular characterization using six ISSR markers to iden… Show more

“…The same product was realized with the monokaryons in the Pleurotus djamor . The dikaryotic mycelium was characterized by the presence of clamp connections and verified under the microscope 10(x) [ 23 ]. Authors have reported that this method for the production of hybrid strains presented a high degree of polymorphism among the hybrid strains and the parental strains.…”

“…When mycelium growth became visible, the whole content of the flask was then homogenized for 30 s and MEA plates were inoculated with 25 or 50 µL of this homogenate and incubated at 28 °C. Developing colonies were microscopically (10x and 40x) examined to identify monokaryotic colonies, i.e., without clamp connections (Valencia del Toro and Leal-Lara, 1999;Sánchez-Hernández et al, 2019). Such colonies were isolated by transferring to new MEA plates and further evaluated for absence of clamp connections, and then considered as neohaplonts.…”

“…To evaluate compatibility, a piece of agar 0.5 cm 2 was cut from a full-grown culture and placed together on a MEA plate with another piece of agar with mycelium from another culture and then incubated at 28 °C. Clamp connections, indicating different compatibility types, were detected by daily inspection under the microscope (10x y 40x) (Valencia del Toro and Leal-Lara, 1999;Sánchez-Hernández et al, 2019). By this way, the two neohaplont types were identified for each dedikaryotized P. eryngii strain, FQ and MB.…”
