Characterization of Plasmid Deoxyribonucleic Acid from Neisseria gonorrhoeae

Abstract: Five of six independent gonococcal isolates have been found to harbor a plasmid pool of covalently closed-circular deoxyribonucleic acid about 2.4 x 106 daltons in size. The plasmid species were 0.5-mol fraction of guanine plus cytosine and comprised 6 to 8% of the total gonococcal deoxyribonucleic acid equivalent, to approximately 24 to 32 copies per cell. There was no apparent correlation between the presence of these plasmid species and the in vitro clonal variation of gonococci associated with the loss of … Show more

“…Engelkirk and Schoenhard (10) found only 33% V colony types after 11 medium. Mayer et al (20) incubated their cultures for 12 to 16 h, but did not report the fraction of piliated cells from which plasmid DNA was isolated.…”

“…Sucrose gradient centrifugation. Five-milliliter linear 20 to 31% sucrose gradients made in either 1 M NaCl, 0.05 M Na2 ethylenediaminetetraacetic acid, pH 7.5, or 1 M NaCl, 0.01 M Na2 ethylenediaminetetraacetic acid, and 0.3 N NaOH, pH 12.5, were used to characterize the CCC DNA isolated from these gonococcal strains. The reference plasmid was [3H Ithymidine-labeled Col El DNA, isolated from Escherichia coli JC 411, which has sedimentation coefficients of 23S and 17S, for the CCC and open-circular molecules, respectively, in neutral sucrose gradients (3).…”

“…We have analyzed these strains for their plasmid content. All three of them carry the common cryptic 4.2-kb plasmid (14), two contain a large 38-kb plasmid (24), and all three carry a third plasmid species of 7.4 kb. The Ampr phenotype is due to this 7.4-kb plasmid.…”

“…To our knowledge, this is the first report describing the presence of a maintenance system on a RCR plasmid. It has been estimated that the copy numbers of RCR plasmids, pTD1 and pJD1, are in the range of 20-25 (Mayer et al, 1974;Ivic et al, 1991), similar to the copy number of pVT736-1 (unpublished results). These findings evoke the following questions.…”
