Although Phragmites australis is commonly planted in constructed wetlands, very little is known about its roots-associated bacterial communities, especially in wetlands used for the remediation of oil produced waters. Here, we describe the bacterial diversity, using molecular (illumina MiSeq sequencing) and cultivation techniques, in the rhizosphere soils of P. australis from an oil-polluted wetland in Oman. The obtained isolates were tested for their plant-growth promoting properties. Most sequences belonged to Proteobacteria, Bacteriodetes and Firmicutes. Sequences of potential hydrocarbondegrading bacteria (e.g. Ochrobactrum, and Pseudomonas) were frequently encountered. All soils contained sequences of known sulfur-oxidizing (e.g. Thiobacillus, Thiofaba, Rhodobacter and Sulfurovum) and sulfate-reducing bacteria, although the latter group made up only 0.1% to 3% of total sequences. The obtained isolates from the rhizosphere soils were phylogenetically affiliated to Serratia, Acinetobacter, Xenorhabdus, Escherichia and Salmonella. All strains were able to solubilize phosphate and about half were capable of producing organic acids and 1-aminocyclopropane-1-carboxylate (ACC) deaminase. Around 42% of the strains had the ability to produce indole acetic acid and siderophores. We conclude that the rhizosphere soils of P. australis in oil polluted wetlands harbor diverse bacterial communities that could enhance the wetland performance through hydrocarbon degradation, nutrient cycling and supporting plant growth.

Yeast Associated with Rice Phylloplane and Their Contribution to Control of Rice Sheath Blight Disease

The phylloplane is an important habitat for yeasts and these yeasts may have antagonistic activities against pathogens and could be used as biocontrol agents. To investigate rice phylloplane yeasts, 282 strains were isolated from 89 rice leaf samples and identified as 15 known yeast species in the phylum Ascomycota and 35 known and two potential new species in the phylum Basidiomycota. The majority of rice phylloplane yeasts belonged to the phylum Basidiomycota. The evaluation of antagonistic activities of 83 yeast strains against rice pathogenic fungi Pyricularia oryzae, Rhizoctonia solani, Fusarium moniliforme, Helminthosporium oryzae and Curvularia lunata revealed that 14 strains inhibited these pathogens. Among the antagonistic strains, Torulaspora indica DMKU-RP31, T. indica DMKU-RP35 and Wickerhamomyces anomalus DMKU-RP25 inhibited all rice pathogens tested, and the production of volatile organic compounds, fungal cell wall degrading enzymes and biofilm were the possible antagonistic mechanisms against all rice pathogens tested in vitro. These yeast strains were evaluated for controlling rice sheath blight caused by R. solani in rice plants in the greenhouse and were found to suppress the disease by 60.0–70.3%, whereas 3% validamycin suppressed by 83.8%. Therefore, they have potential for being developed to be used as biocontrol agents for rice sheath blight.

Nitrogen, P and K are essential macronutrients that are not readily available to plants. Rhizobacteria are able to convert these unavailable forms for subsequent uptake by the plant, diverse species have been characterized as N2 fixers, P solubilizers and capable to solubilize mineral K from unavailable forms. The objective of this study was to select rhizobacteria capable of improving NPK availability and promoting tomato (Solanum lycopersicum L.) growth. Fifteen strains were studied. Four strains were selected for their capacity to fix N2, three for their ability to solubilize P, and six for their capacity to solubilize biotite and K-feldspar, isolated from tomato, lentil, chili pepper, faba bean and lettuce crops in Andisol and Alfisol soils. Through 16SrRNA sequencing, selected strains were identified as Pseudomonas gessardi, P. koreensis, P. brassicacearum, P. marginalis, Acinetobacter calcoaceticus and Rahnella aquatica. Phosphorus solubilizing strains did not show a positive effect on plant growth or an increase in available soil P. The N2 fixing bacteria Tmt-16 strain increased root growth in 23.57%; maintained the highest N content in plant tissue, 2.60%, higher amount of N available in the soil, 2.95 mg kg-1 , and a higher content of N-NH4 + 1.95 mg kg-1. The K solubilizing strains Ls-C21, Ltj-62 and LsC-58 reached 17.0 to 19.0 mg kg-1 available K and 0.04 to 0.05 mg kg-1 exchangeable K (p ≤ 0.05.). These four endemic rhizobacteria can be potentially used as biofertilizers, allowing a reduction in the use of chemical fertilizers and a more sustainable production of tomatoes.
