Food Research 4 (1) : 85 -90 (February 2020) Journal homepage: AbstractSuji is a plant that has long been used as food colorant and medicinal plant in Indonesia. This study aimed to determine the immunomodulatory activity of fractions obtained from suji leaves by using in vitro phagocytic macrophage assay and also to determine the phytochemical compounds of the most active fraction. Initially, Suji dry leaves powder were macerated with 96% ethanol upon filtration, then the filtrate obtained was evaporated to dryness. The extract obtained was triturated with chloroform to give fraction soluble (F1) and insoluble fraction (F2). Then, F1 was partitioned using a mixture of hexane-methanol-water (25: 14: 1 v /v) to give two layers, namely upper layer (F3) and lower layer (F4). Furthermore, the four fractions (F1, F2, F3 and F4) were tested by in vitro phagocytic macrophages method at the concentration of 10, 25, 50 and 100 µg/mL. The parameters used were the Phagocytic Index (PI) and the Phagocytic Capacity (PC). Macrophage phagocytic in vitro assay showed that fractions of the suji leaf ethanolic extract could improve the phagocytic activity of macrophages. The fraction that demonstrated the highest activity was subjected to phytochemical study by Thin Layer Chromatography (TLC). Based on the data obtained, F3 was the most active fraction, because it has the highest PC and PI values compared to other fractions or negative control. Establishment and characterization of macrophage cell line from thymus ofCatla catla(Hamilton, 1822)

A continuous cell line has been developed from thymus explants of Catla catla and the cells have been subcultured for 63 passages. The cells exhibited optimum growth at 30°C in L-15 medium containing 15% foetal bovine serum. The cultured cells engulfed yeast cells and fluorescent latex beads. These cells produced reactive oxygen and nitrogen intermediates following stimulation with lipopolysaccharide and phorbol esters. The culture supernatant from the cultured cells had lysozyme activity and these cells demonstrated Fc receptors. Almost all the cells were positive for alpha-naphthyl acetate esterase enzyme suggesting that the cells are of macrophage lineage and therefore, the cell line was designated as catla thymus macrophage (CTM) cell line. CTM cells formed aggregates around zoospores of Aphanomyces invadans, but were unable to inhibit the germination of spores. The karyotype analysis of CTM cells at 25th passage revealed a typical diploid model with 50 chromosomes per cell. Partial amplification, sequencing and alignment of fragments of two mitochondrial genes 16S rRNA and cytochrome c oxidase subunit 1 confirmed that the CTM cell line originated from C. catla. This cell line should be useful for studying the role of macrophages in differentiation and maturation of thymocytes and can be a source of macrophage-specific enzymes and cytokines.

Red onion skin (Allium cepa var ascalonicum) contains various ingredients that may function as antibacterial agents against microorganisms, as well as anti-inflammatory and immunomodulator agents for host cells, such as macrophages. Pseudomonas aeruginosa found in the oral cavity is commensal bacteria that may turn into opportunistic pathogen by utilizing its virulence factors such as motility and adhesion to the host cell. The purpose of this study was to investigate the effect of red-onion-skin ethanolic extract towards P. aeruginosa ATCC 9027 on the motility and adhesion ability, furthermore, to know its effect on the macrophage phagocytosis. The research was conducted into three parts of experiment using red-onionskin ethanolic extract. Extract-induced bacterial motility test was carried out on semi-solid media, stained using 0.1% crystal violet, then the radial length of the bacterial movement was measured. The bacterial adhesion index to buccal cells was calculated after incubated for two hours and stained with Gram stain. Phagocytic activity of the host cells on P. aeruginosa was done by exposing the extract to the mouse peritoneal macrophages, then the phagocytosed bacteria were counted after Giemsa staining. Statistical test results from the three experiments showed significant differences between the test groups compared to the control groups (p <0.05). It was concluded that the red onion-skin ethanolic extract not only affects P. aeruginosa by reducing swarming motility and preventing bacterial adhesion to buccal epithelial cells, but also induces the host cells by increasing the ability of macrophage phagocytosis to these bacteria.
