(1) Background: estragole is a monoterpene found in the essential oils of several aromatic plants, which can be used for several pharmacological activities. The aim of this study was to evaluate the antinociceptive effect of estragole (Es) and its β-cyclodextrins inclusion complex (Es/β-CD). (2) Methods: the effects of Es and Es/β-CD on the central nervous system (CNS) were evaluated through open field and rota-rod assays, and the antinociceptive effect in formalin models, abdominal writhing induced by acetic acid, hot plate, tail flick test and plantar mechanical hyperalgesia. (3) Results: Es and Es/β-CD showed no alterations on the CNS evaluated parameters and the results suggested there was an antinociceptive action in the formalin, abdominal writhing, hot plate, tail flick tests and plantar mechanical hyperalgesia, proposing the involvement of the nitric oxide, glutamatergic signaling pathways, cyclic guanosine monophosphate and vanilloid pathways. (4) Conclusion: the results suggest that Es and Es/β-CD have a promising antinociceptive potential as a possible alternative for the pharmacological treatment of pain, also showing that the encapsulation of Es in β-cyclodextrins probably improves its pharmacological properties, since the complexation process involves much lower amounts of the compound, contributing to better bioavailability and a lower probability of adverse effect development.

The search for vegetable extracts for phytosanitary control has been expanded to find new active ingredients to control plant diseases. This study aimed to evaluate the in vitro effect of the fixed constituents of Copaifera luetzelburgii, Croton zehntneri and Lippia lasiocalycina, at the concentrations of 2, 20, 200 and 2,000 μg mL-1, on the percentage of mycelial growth inhibition of Colletotrichum siamense, C. truncatum, Fusarium sacchari, F. udum, Lasiodiplodia theobromae and Thielaviopsis ethacetica, as well as the conidium concentration of C. siamense, F. sacchari and F. udum produced in culture medium with all the extracts. The tested ethanolic extract, especially at the highest concentration, inhibited the percentage of mycelial growth and/or conidium concentration of the evaluated fungi. The other concentrations showed low inhibitory effects or no activity against the fungi. The average values for percentage of mycelial growth inhibition of the ethanolic extract from L. lasiocalycina, C. zehntneri and C. luetzelburgii against the six fungi were 62.5, 53.4 and 51.0 %, respectively. The ethanolic extract of L. lasiocalycina showed the most significant effect on the percentage of mycelial growth inhibition and conidia concentration. The fixed constituents of C. luetzelburgii, C. zehntneri and L. lasiocalycina at 2,000 μg mL-1 showed to be efficient in inhibiting the mycelial growth of C. siamense, C. truncatum, F. sacchari, F. udum, L. theobromae and T. ethacetica, and inhibit the conidia production of C. siamense, F. sacchari and F. udum.

Cyclodextrins are nanometric cyclic oligosaccharides with amphiphilic characteristics that increase the stability of drugs in pharmaceutical forms and bioavailability, in addition to protecting them against oxidation and UV radiation. Some of their characteristics are low toxicity, biodegradability, and biocompatibility. They are divided into α‐, β‐, and γ‐cyclodextrins, each with its own particularities. They can undergo surface modifications to improve their performances. Furthermore, their drug inclusion complexes can be made by various methods, including lyophilization, spray drying, magnetic stirring, kneading, and others. Cyclodextrins can solve several problems in drug stability when incorporated into dosage forms (including tablets, gels, films, nanoparticles, and suppositories) and allow better topical biological effects of drugs at administration sites such as skin, eyeballs, and oral, nasal, vaginal, and rectal cavities. However, as they are nanostructured systems and some of them can cause mild toxicity depending on the application site, they must be evaluated for their nanotoxicology and nanosafety aspects. Moreover, there is evidence that they can cause severe ototoxicity, killing cells from the ear canal even when applied by other administration routes. Therefore, they should be avoided in otologic administration and should have their permeation/penetration profiles and the in vivo hearing system integrity evaluated to certify that they will be safe and will not cause hearing loss.
