Characterization of hydrochloride and tannate salts of diphenhydramine

Abstract: Proper characterization is an important aspect of any dosage form design. The objective of this work was to characterize tannate salt and hydrochloride salt of diphenhydramine. As a part of characterization studies, Differential scanning calorimetry was used to investigate thermal effects and nature of salts, supported by X-ray powder diffraction. Scanning electron microphotographs was used to surface topography of salts of diphenhydramine. Fourier-transform infrared spectroscopy, solubility study and flowabil… Show more

“…The spectrum is found to be similar with For DPH, the peaks in the range of 2,400-2,700 cm −1 are attributed to the stretching vibrations of -N(CH 3 ) 2 ·HCl bond, while the peak at 1,103 cm −1 represents the O-CH 3 stretching vibration (24). No peaks in the range of 2,400-2,700 cm −1 are observed in the matrix-drug blends, implying the deformation of ionic bonds between hydrochloric acid and dimethylamino, when DPH be loaded in the matrix.…”

“…8). The first and third was assigned to C-H out-of-plane bending vibration of monosubstitutes phenyl on a few studies [36,37], while the latter two were assigned to C-H out of plane deformation of monosubstituted phenyl [3]. The same three wavenumbers were found for the physical mixture although the peaks were much smaller.…”

“…DPH belongs to the class of ethanolamine H1 receptor antagonist and possesses a significant anticholinergic effect in addition to antihistaminic activity [3]. It is produced under the trade name Benadryl, and commonly used motion sick pills also contain DPH, although in combination with 8-chlorotheophylline to reduce the drowsiness created by DPH.…”
