Characterization of Class-3 Semaphorin Receptors, Neuropilins and Plexins, as Therapeutic Targets in a Pan-Cancer Study

Abstract: Class-3 semaphorins (SEMA3s), initially characterized as axon guidance cues, have been recognized as key regulators for immune responses, angiogenesis, tumorigenesis and drug responses. The functions of SEMA3s are attributed to the activation of downstream signaling cascades mainly mediated by cell surface receptors neuropilins (NRPs) and plexins (PLXNs), yet their roles in human cancers are not completely understood. Here, we provided a detailed pan-cancer analysis of NRPs and PLXNs in their expressio… Show more

“…With the continued exploration of lncRNAs (Zhang et al, 2018a) and the development of RNA-centric approaches (Zhang et al, 2018b), we will be able to identify the interaction targets of lncRNAs in a native context (Zhang et al, 2020). The identification of these targets will further improve our understanding of lncRNA-mediated pathway regulation and advance our understanding of lncRNA functions (Zhao et al, 2019).…”

“…In the research on tumors, on the one hand, PLXNC1 was identified as a protective gene in acute myeloid leukemia, papillary thyroid cancer, and melanoma [42][43][44]. On the other hand, PLXNC1 is considered to be related to the poor prognosis of gastric cancer and liver cancer [45,46], and was mainly related to the aggressive phenotype in a pan-cancer study [47]. There are few studies on PLXNC1 in NB.…”

“…Further, there is a collaboration between c-Met and several co-receptors, e.g., α6β4 integrin [148], as well as between plexin A/B with NRP-1/NRP2 [43,96] and L-CAM [149]. The interaction of plexin A with NRPs has recently been described in detail [150]. Initially, an interaction is started between a plexin A glycoprotein and a neighboring NRP-1/NRP-2.…”
