Oneand two-dimensional gel electrophoresis and cellular immunoblotting were used to compare the protein profiles and immunogenic capabilities of salt-and sodium dodecyl sulfate-extracted components isolated from different BruceUla species. Cellular immunoblotting demonstrated that freshly isolated bovine peripheral blood mononuclear cells proliferated to similar molecular mass components from the various Brucella extractions. One-and two-dimensional gel electrophoresis showed similarities in protein profiles among the different BruceUla species that correlated with lymphocyte reactivity. The results presented in this study provide preliminary evidence that common proteins that induce lymphocyte proliferation are present among different Brucella species, suggesting that a genuswide subunit vaccine may be feasible.

Journal homepage: http://www.ijcmas.com Brucellosis is an important zoonotic disease globally that causes huge economic losses to the livestock owners and is of public health significance. Brucellosis in animals is endemic in India. In sheep and goats, Brucellosis is mainly caused by Brucella melitensis whereas Brucella ovis causes the disease in sheep. The symptoms in infected sheep and goats are abortions, stillbirths and the birth of weak offsprings. Animals that abort may retain the placenta. Sheep and goats usually abort only once, but reinvasion of the uterus and shedding of organisms can occur during subsequent pregnancies. Several studies have been carried out on seroepidemiology of caprine and ovine Brucellosis in India. The tests commonly used for diagnosis of Brucellosis are the milk ring test, Rose Bengal Plate Test (RBPT), Standard Tube Agglutination Test (STAT), Microtiter Plate Agglutination Test (MAT) and ELISA. The RBPT is a rapid screening test for the diagnosis of Brucellosis. The sensitivity of RBPT is very high (>99%) but the specificity can be low and it could sometimes give a false positive result. Its positive predictive value is low and a positive test result requires confirmation by a more specific test. Summary A study was conducted to evaluate the biological activity of Brucella allergens extracted with hydrochloride or trichloroacetic acid. Smooth and mucoid Brucella abortus cells and the medium in which brucellae were propagated were used to prepare the allergens. The biological activity of the allergens was estimated in guinea‐pigs sensitized with Brucella abortus strain 544. The guinea‐pigs were intradermally injected with several allergen dilutions. The dilutions were coded and randomized for site of injection so that none of the dilution was injected twice on the same site. Variance analysis using incomplete Latin square was used for the statistical calculation of the results. The calculated biological activity of the allergens was compared with the biological activity of a ‘standard’ allergen that has proved effective in detecting cattle brucellosis. The skin erythema diameter was best when recorded 32 h after allergen injection. Statistical analysis of the skin erythema diameters showed a great variation in biological activity (12–105%) between the allergens. Only the allergen extracted from the medium in which a mucoid Brucella strain was propagated was as potent as the standard. The use of the incomplete Latin square for variance analysis resulted in the estimation of the biological activity of nine batches of allergen in only 27 guinea‐pigs.
