There is a great deal of research interest regarding substitutes for antibiotics because of various obstacles to the efficacy and use of antibiotics. We isolated and analyzed diversity of microbiota which exhibited antibacterial activity against 23 pathogenic bacteria, to develop alternative agent of antibiotics. By investigating the microbiota from rocks on the seashore, we characterized and obtained various antibacterial material-producing bacteria. Thirty-one isolates belong to four genera and seven species, according to 16S rDNA sequence analysis, showed antibacterial activities against 23 pathogenic bacteria. The Identity of 16S rDNA sequences indicated three species of Bacillus, one species of Paenibacillus, one species of Pseudomonas and two species of Enterobacter. Two isolates were similar to Bacillus aerophilus, four isolates were similar to Bacillus pumilus, seven isolates were similar to Bacillus safensis, 15 isolates were similar to Paenibacillus polymyxa, respectively. In addition, one isolate was similar with Pseudomonas poae, one isolate was similar to Enterobacter asburiae, and one isolate was similar to Enterobacter ludwigii, respectively. Variations of antibacterial activity and level among the same species were indicated the diverse strains of isolates. Vibrio vulnificus showed the highest degree of growth inhibition by 29 isolates. Further studies regarding antibacterial materials and bacteria suggest that development of probiotic strains or alternative agents to antibiotics.

Agricultural and livestock microorganisms including bacteria, yeast and fungi have been used as eco-friendly biological agent in plant growth promoting, disease suppression and odor reduction. In this study, we have optimized culture conditions of three species, such as Bacillus subtilis, Saccharomyces cerevisiae and Lactobacillus casei that could act as effective biological agent by producing growth-promoting factors to crops. They also reduce harmful odors generated from livestock manure. We made up the optimal media recipes for Bacillus subtilis, Saccharomyces cerevisae and Lactobacillus casei named as MMS (glucose 0.5%, soy bean flour 0.8%, NaCl 0.15%, K 2 HPO 4 0.25%, Na 2 CO 3 0.05%, MgSO 4 0.1%), YSM (dextrose 1%, soytone 0.5%, yeast extract 0.5%) and LSM (glucose 2%, soytone 0.5%, casein milk 0.5%, NaAc 0.5%, KH 2 PO 4 0.2%, MgSO 4 0.002%, MnSO 4 0.05%), respectively. The culture period of three microorganisms were determined on 30 hours in 500 L fermenter. In addition, the maximal cell growth of Bacillus subtilis, Saccharomyces cerevisae and Lactobacillus casei was measured as 1.4×10 9 cfu/mL, 6.4×10 8 cfu/mL, 3.3×10 9 cfu/mL, respectively. From above results, we represent that development of practical media and cultural techniques can economize heavily the cost for mass production of agricultural and livestock microorganisms.

The Antibacterial Activity Against Fish Pathogen of Paenibacillus sp. MK-11 Isolated from Jeju Coast

In this study, we isolate and identify bacteria from seawater collected from Jeju coast, to evaluate the antimicrobial activity against the fish pathogenic bacteria. 14 bacterial strains were isolated and identified using physiological, biochemical and molecular tools. Antibacterial activity of all the 14 isolates were screened against four major fish pathogens namely, two Gram-positive: Streptococcus iniae, Streptococcus parauberis and two Gram-negative: Vibrio anguillarum, Edwardsiella tarda. Results revealed that among the 14 isolates, MK-11 was found to have antibacterial activity against S. iniae, S. parauberis, V. anguillarum Particularly, S. iniae was susceptibility with the MIC value of 250 μg/ml. The biochemical and physio-chemical results reveal that MK-11 had the sugar-alcohol disassemble ability of the D-sorbitol and D-mannitol. Also the utilization of the yeast extract, sorbitol and di-potassium phosphate were noted to be high. The optimum culture condition such as pH and temperature was recorded as pH 6.0, 25℃ and along with 1% NaCl which differs from the previous reports particularly in nutrient resolutions. As results of the analysis of 16S rDNA sequences, MK-11 show the high similarity with Paenibacillus polymyxa, P. jamilae, P. brasilensis 99.78%, 99.43%, 99.39%, repectively. Hence, in the present study, the isolated Paemibacillus sp. MK-11 from Jeju seawater possesses the antibacterial activity against fish pathogens and it could be used as a new antibiotic agents against the gram positive fish pathogens.
