Characterization of an anti‐varicella‐zoster virus compound that targets the portal protein encoded by ORF54

Abstract: An anti-varicella-zoster virus compound, a 5-chlorobenzo[b]thiophen derivative (45B5), was characterized. Its 50% effective concentration against the cell-free vaccine Oka strain and 50% cytotoxic concentration in human fibroblasts were 16.9 µM and more than 100 µM, respectively. Treatment with 45B5 decreased viral DNA synthesis and IE62 expression weakly but significantly. All 45B5-resistant viral clones isolated were found to have at least one mutation in ORF54 that encodes the portal protein. There were no … Show more

“…The large terminase subunit has an ATPase activity that provides energy for packaging initiation and termination [36],[37]. Portal protein (encoded by ORF77) shares high amino acid sequence similarity with Staphylococcus phage vB_SepS_SEP9, which can inject DNA into the host cell through a pathway formed by portal protein [38]. Assembly protease (encoded by ORF78) and DNA packaging protein (encoded by ORF81) were also found in phage SA2.…”

“…phi-pp2. ORF181 was predicted to encode a portal protein with 100% identity to that of Vibrio phage phi-pp2, which can infect DNA into the host cell through a pathway formed by portal protein (Yasui et al, 2017). Just as reported (Paul et al, 1998), terminase plays an important role in the DNA packing processes of the phage genome.…”

“…Packaging related proteins: the terminase is a packaging protein that inserts a single phage genome within an empty capsid structure, typically consisting of one large and one small subunit, the small subunit speci cally recognizes viral DNA and attracts the large subunit for initial cleavage, the large subunit has ATPase activity and can provide energy for phage packaging initiation [26]. Phage portal protein can form a pathway to inject DNA into the host and participate in the protein connection between the phage head and tail [29].…”
