Characterization of a xylanolytic bacterial strain C10 isolated from the rumen of a red deer (Cervus elaphus) closely related of the recently described species Actinomyces succiniciruminis, A. glycerinitolerans, and A. ruminicola

Abstract: Gram-stain-positive, catalase and oxidase-negative and short rod-shaped bacterium C10 with occasional branching was isolated under strictly anaerobic conditions from the rumen fluid of a red deer (Cervus elaphus) in the course of study attempting to uncover new xylanolytic and cellulolytic rumen bacteria inhabiting the digestive tract of wild ruminants in the Czech Republic. The anaerobic M10 medium containing bovine rumen fluid and carboxymethylcellulose as a defined source of organic carbon was used in the p… Show more

“…Exocellulases appear to be provided by a taxonomically narrow group of organisms including Ruminococcus, OPB54 Rhizobiales, and Actinomycetales. These bacteria have been identified as lignocellulose degrading organisms in a variety of environments and likely represent the true cellulolytic consortium of the enteric microbial community despite comprising a relatively small fraction of the overall community (<1–17%) (Ben David et al, 2015; Joshi et al, 2018; Šimůnek et al, 2018).…”

“…The presence of such a variety numbers of GHase belongs to various GHase families that were not reported earlier in the P. stutzeri genome [ 35 ]. The enzyme products of GH1, and GH30 families having specific role in the processing of xylan to xylose which are plant based oligosachharides, hence their presence into the genome of MP4687 presume the role of this bacteria into biomass hydrolysing and the rumen functioning of cattle [ 36 , 37 ]. It is assumed that many of the genes associated with these specific functions were transferred from other ruminal bacterial species to MP4687, to confer additional power to survive in the rumen environment [ 29 ].…”

“…Fibrobacter succinogenes is a major contributor to cellulose digestion in the rumen of cattle [ 37 ]. It is capable of breaking down many sugars to produce formate, acetate, and succinate [ 38 ]. All these results were concordant with that the supplementation of 1,200 mg/kg NA elevated the concentrations of acetate, butyrate, and TVFA in the rumen.…”
