Characterization of a Dengue Patient Cohort in Recife, Brazil

Abstract: From 2004 to 2006, 658 patients with suspected dengue virus infections were enrolled in a clinical dengue cohort established in Recife, Pernambuco, located at the northeastern region of Brazil. A total of 2,364 blood samples were collected, and serum, plasma, and cells were cryopreserved. Among the suspected cases, 354 (54%) were confirmed as acute DENV-3 infection based on reverse transcription-polymerase chain reaction, virus isolation, and ELISA-IgM. According to WHO criteria, 29.4% of the positive acute ca… Show more

“…This product was found to have high anti-dengue antibody titers of 1:1280 for DENV-2 and 1:160 for DENV-1 and DENV-3. This was expected, since this IVIG was prepared using blood donations from Brazil, which is an endemic area (Cordeiro et al, 2007). In Brazil, three serotypes are currently in circulation, DENV-1, 2, and 3, while infection by DENV-4 is more restricted to the North Region (Roraima, Manaus and Belém) (Brazil, 2010(Brazil, , 2011a(Brazil, , 2011b.…”

“…The risk of death, in this group, is associated to the difficulty in handling the disease in a population with high frequency of comorbidities 30 . In addition to that, in areas where the occurrence of dengue is relatively new, such as in Minas Gerais and in most part of Brazil 31 , older people are not entirely immune and therefore, they have an increased risk of secondary infection 32 and, consequently, greater risk of a more severe disease. In Brazil, the co-circulation of viral serotypes of dengue is frequent, with alternated predominant serotypes.…”

This maternal anti-dengue immune profile seems to be different from studies conducted in pregnant women in Asian countries where the majority of pregnant women were immune against two or more DENV serotypes 5,8,9 .…”

“…Dengue cases tracked in febrile infants were laboratory-confirmed by reverse-transcriptase polymerase chain reaction (RT-PCR) and IgM-ELISA. RT-PCR was performed according to a standardized protocol described elsewhere 17 . Viral RNA was extracted from serum samples using QIAquick PCR purification kits (Qiagen, Valencia, USA).…”
