Characterization of a Cell Culture Adapted Goat Pox Virus¹

Abstract

Summary An attenuated goat pox virus (GPV) in sheep testicle cell (STC) culture was further adapted to sheep embryo dermis cell (SEDC) culture and was characterized in terms of its growth pattern and its resistance to physical and chemical treatments. A single growth curve in SEDC showed an eclipse period of less than 18 hrs, followed by exponential growth with its maximum at 4 days post‐infection. Thereafter, a plateau was established till 11 days post‐infection with an insignificant fall in virus titre occurring after 9 days post‐infection. In the following two weeks of incubation the total fall in titre slightly exceeded 1 log. Intracellular virus was much higher than extracellular virus during the exponential growth; it did remained higher up to 18 days post‐infection, when intra‐ and extracellular virus were equal in amount. The strain was resistant to heat: 450 and 30 minutes, respectively, were needed to inactivate the strain at 50 °C and 56 °C. Chloroform and ether caused complete inactivation of the strain. Trypsin caused gradual inactivation of the attenuated GPV, but complete inactivation was not achieved before 2 hours of treatment with the proteolytic enzyme in association with ethylenediamine‐tetraacetic acid (EDTA). Zusammenfassung Charakterisierung eines Zellkultur‐adaptierten Ziegenpockenvirus Ein in Schafhoden‐Zellkulturen (STC) attenuiertes Ziegenpockenvirus (GPV) wurde anschließend an embryonale Schafhaut‐Zellkulturen (SEDC) adaptiert und in bezug auf sein Ver‐mehrungsverhalten und seine Widerstandsfähigkeit gegenüber physikalischen und chemischen Behandlungen charakterisiert. Die Einschrittvermehrungskurve in SEDC ließ eine Eklipsephase von weniger als 18 Stunden mit anschließender exponentieller Vermehrung des Virus erkennen, die in einem Maximum am 4. Tag p. inf. gipfelte. Danach stellte sich ein Plateau ein mit einem nicht signifikantem Titerabfall am 11. Tag. In den darauffolgenden 2 Wochen Inkubation erreichte die Reduktion des Virustiters etwas mehr als 1 log 10. Während der exponentiellen Vermehrung war der Gehalt an intrazellulärem Virus viel höher als an extrazellulärem; er blieb bis zum 18. Tag p. inf. höher und war dann dem extrazellulären Gehalt gleich. Der Stamm verhielt sich wärmeresistent: zur Inaktivierung waren 450 bzw. 30 min Inkubation bei 50 bzw. 56 °C notwendig. Chloroform und Äther inaktivierten den Stamm restlos. Trypsin bewirkte eine nur graduelle Inaktivierung des attenuierten GPV. Durch eine Behandlung mit diesem proteolytischen Enzym in Verbindung mit Äthylendiamintetraessigsäure (EDTA) ließ sich eine vollständige Inaktivierung nicht unter 2 Stunden erreichen.
