Characteristics of proteoglycans isolated from small and large bovine ovarian follicles.

Abstract: Acetone precipitates of bovine follicular fluid from small (3-5 mm) and large (10-20 mm) follicles were fractionated by nondissociative procedures involving chromatography on Sephacryl S-300, DEAE cellulose, and Sepharose CL-2B. Most of the proteoglycan material eluted with a relatively low molecular weight (Kav = approximately 0.6) from Sepharose CL-2B. Comparisons of the proteoglycans from small versus large follicles showed respective differences of 20 versus 34% protein, 74 versus 48% chondroitin sulfate-B… Show more

“…These proteoglycans and their glycosaminoglycan side chains are osmotic solutes, which act to increase the osmotic pressure inside of the follicle, resulting in fluid accumulation (Grimek and Ax 1982;Bellin et al 1983;Grimek et al 1984;Clarke et al 2006). In accordance with the present results, Vasconcelos et al (2012) also demonstrated that the presence of GDF-9 in culture medium of bovine preantral follicles was not accompanied by an enhancement of the number of follicles showing antrum formation.…”

“…The CS/DS GAGs were mostly 4-sulfated. This is also the case in bovine follicular fluid GAGs [6], while porcine follicular fluid contains unsulfated, 4-sulfated, and 6-sulfated CS/DS GAG chains [2], and human follicular fluid contains both unsulfated and 6-sulfated GAGs. Thus there is clearly species variation in the level and type of sulfation of the GAGs in follicles.…”

“…Proteoglycans are therefore a diverse range of molecules with diverse functions, documented to be involved in cell growth and Glycosaminoglycans have been identified in the ovarian follicular fluid of pigs [2][3][4], cows, [5][6][7][8][9], humans [10][11][12], and rats [13,14]. The predominant GAGs in bovine and porcine follicular fluid are dermatan sulfate (DS) and chondroitin sulfate (CS) [2,6]. The CS/DS-containing GAGs were shown to be attached to a protein core in bovine follicles by Grimek and Ax [9], while the GAG heparan sulfate (HS) was observed not to be bound to a protein core.…”

“…Hyaluronan is a major glycosaminoglycans present in the cervical mucus, the cumulus, follicular fluid, and seminal plasma (6)(7)(8). This macromolecule, also present in the oviduct and uterine fluids, increases up to the time of implantation (9).…”
