Characteristics of Five New Photoautotrophic Suspension Cultures Including Two Amaranthus Species and a Cotton Strain Growing on Ambient CO2 Levels

Abstract: Suspension cultures of cotton (Gossypium hirsutum), Amaranthus cruentus, A. powellii, Datura innoxia, and a Nicotiana tabacum-N. glutinosa fusion hybrid were adapted to grow photoautotrophically under continuous light. The cotton strain grew with an atmosphere of ambient CO2 (about 0.06 to 0.07% in the culture room) while the other strains required elevated CO2 levels (5%). Photoautotrophy was indicated by the requirement for CO2 and for light for growth. The strains grew with doubling times near 14 days and h… Show more

“…The cotton and soybean photoautotrophic lines have more rapid doubling times than the other lines, averaging 6 to 7 d for COT-62, 7 d for COT-P, and 8 to 9 d for doubling ofboth SB 1-P and SB-P. A longer doubling time of 12 d was found for DAT-P, NTG-P, and APO-P. The cells cultured with MIN medium do not differ in doubling time from those cultured on KTP°or KN°me-dium (22,26).…”

“…In the previous studies with photosyn- proposed that the low levels of RuBP carboxylase present were analogous to the low levels observed in developing leaves (13,32). The reason for the difference in RuBP carboxylase levels in the different cell lines is unknown, but it is evident that all four of the cell suspensions studied in detail here are much more like cells in mature leaves.…”
