Objectives: This study was conducted to assess the prevalence and characterization of Staphylococcus aureus from chicken and quail eggshells and to study the antibiogram of the isolates. Materials and methods: A total of 300 eggs (220 chicken eggs and 80 quail eggs) were collected from different retail shops and farms in Mymensingh district. Swabs taken from the egg surfaces were cultured on Mannitol Salt Agar for the isolation of S. aureus . Polymerase chain reaction was conducted for confirmatory identification of the bacterial species targeting nuc gene, followed by confirmation of methicillin-resistant S. aureus by targeting the mecA gene. Antibiotic sensitivity test of the isolated bacteria was done against commonly used antibiotics by the disk diffusion method. Results: The prevalence of Staphylococcus spp. and S. aureus in the chicken eggshell surface was 20.45% and 10.45%, respectively. Similarly, the prevalence of Staphylococcus spp. and S. aureus in quail eggshell surface was 16.25% and 5%, respectively. Overall, 27 isolates were identified as S. aureus , of which 23 were from the chicken eggshell surface and four from quail eggshell surface. Among the seven isolates tested, overall four (57.14%) were positive for the nuc gene. On the other hand, the mecA gene could be detected in three (50%) S. aureus out of six oxacillin resistant isolates. The antibiogram study indicated that most of the isolates were resistant to the antibiotics under β-lactam group. Conclusion: The present study concludes that chicken and quail egg surface harbor multidrug-resistant bacteria which may cause public health hazards, if these antibiotic-resistant bacteria are transferred to a human.

ARTICLE INFOThis study was conducted to record the potential risk of some pathogens in table eggs in Beni-Suef city, Egypt. A total of 100 table eggs samples (Farm and Baladi eggs) were randomly collected from poultry farms, markets, supermarkets and groceries in Beni-Suef city, 50 farm eggs samples (each of 3 eggs) and 50 baladi eggs samples (each of 5 eggs) were examined for the presence of coliforms, E.coli, Salmonella, coagulase positive Staphylococcus aureus and Staphylococcal enterotoxins. Isolates were identified by biochemical, serological and molecular (PCR) methods. The obtained results in the present study revealed that (22%) of the examined samples were contaminated with Coliforms. The other identified genera were Citrobacter freundii, Citrobacter diversus, Edwardsiella tarda, Enterobacter spp., Morgonella morganii, Klebsiella oxytoca, Providencea spp., Serratia fonticola and Yersinia intermedia. E.coli (atypical type) was detected in a percentage of 27.27%. True fecal E. coli and Salmonella spp. failed to be detected in any of the whole examined 100 eggs samples. Additionally, Staphylococcus spp. was detected with incidence rate of (13%), out of them, 8 (61.5%) isolates were accounted for coagulase positive Staphylococcus aureus. Enterotoxin profiling revealed that two classical enterotoxin genes (SEA and SED) were detected either singly or in combination. The potential health hazards and the suggested control measures of the isolated strains had been discussed.

The aim of the study was to identify the affinity of 10 Staphylococcus strains isolated from table chicken eggs to specific species. Preliminary analysis performed by API ID32 Staph test identified these strains as S. aureus, but they exhibited a negative reaction in the tube coagulase test. Thus, the analysed strains were initially characterised as Staphylococcus aureus-like (SAL). Further characterisation was performed by genotypic methods, using restriction fragment length polymorphism (RFLP) of the coagulase gene (coa) and sequencing of the gene rpoB. An attempt was also made to identify the isolated Staphylococcus strains by MALDI-TOF mass spectrometry. The results indicated that none of the strains tested belonged to the species S. aureus. The rpoB sequences of five isolates showed the highest sequence similarity to S. haemolyticus, three isolates to S. chromogenes, and one isolate to S. epidermidis. One strain (SAL4) remained unidentified in this analysis. The results obtained using mass spectrometry were comparable to those based on gene sequence analysis. Strain SAL4, which could not be identified by sequencing, was identified by MALDI-TOF as Staphylococcus chromogenes.
