Characterisation of a highly sensitive troponin I assay and its application to a cardio-healthy population

Abstract: This new hs-TnI assay is able to measure to an order of magnitude lower than the current generation TnI assay from the same manufacturer. With TnI being detectable in nearly all apparently healthy subject samples this suggests that TnI presence does not always indicate cardiomyocyte necrosis.

“…The 99th percentiles were 9.4, 10.5, and 10.2 ng/L, respectively, in boys and 8.2, 10.1, and 7.9 ng/L in girls. In comparison to adults using the same assay, 99th percentiles were 13.6 ng/L for all individuals, 14.0 ng/L in men, and 11.1 ng/L in women (40 ). Further research will be required to assess the clinical significance or need to report separate 99th percentile values in healthy children/adolescents.…”

“…For hs-cTnI analysis, samples that had been thawed and refrozen once in 2008 and subsequently stored at Ϫ20°C were shipped on dry ice to Akershus University Hospital, Lørenskog, Norway. Prior studies have suggested that hs-cTnI concentrations are not affected by long-term storage or repeated freeze-thaw cycles (15,16 ). Before analysis, thawed samples were centrifuged at 3000 relative centrifugal force for 30 min.…”

“…The long-term stability of cTnI with both the Beckman Coulter high sensitivity (hs)-TnI (14 ) and the Abbott ARCHITECT STAT hs-cTnI assays (15 ) has been demonstrated under these conditions. Assays were performed in singlicate.…”
