Changing Viral Susceptibility of a Human Cell Line in Continuous Cultivation

Abstract: During its serial transfer and cultivation in this laboratory, a human conjunctival cell line (Chang) was observed to change in morphology. Concurrently no change was noted in the susceptibility of the cells to viruses capable of infecting the original cell line. However, it was noted that the derived variant cell line had acquired susceptibility to the induction of cytopathic effects and incomplete virus formation by several strains of influenza viruses. It was then discovered th… Show more

“…Confluent stock cultures of established epithelial cells [HeLa, FL, Y-1 adrenal, human intestine (ATCC strain CCL-6), human conjunctiva (clone 1-5C-4) (25)] were seeded at a concentration of approximately 2.0 X 105 per plastic dish (35-mm) and the resulting monolayers were incubated at 37 C for 18 hr in an atmosphere of 5% CO2 in MEM medium with 5% fetal calf serum. For scanning electron microscopic observation, epithelial cells were seeded on cover glasses placed in a plastic dish.…”

“…Hemagglutination (HA), neuraminidase (NA) assay, complement-fixation (CF) test and infectivity titration were described previously (12). Clone 1-5C-4 cells were grown in medium 199 containing 10% fetal calf serum (23).…”
