Changing expression profiles of lncRNAs, mRNAs, circRNAs and miRNAs during osteoclastogenesis

Abstract: Bone is a dynamic organ continuously undergoing shaping, repairing and remodeling. The homeostasis of bone is maintained by the balance between osteoblastic bone formation and osteoclastic bone resorption. Osteoclasts (OCs) are specialized multinucleated cells derived from hematopoietic stem cells (HSCs) or monocytes/macrophage progenitor cells. There are different stages during osteoclastogenesis, and one of the most important steps to form functional osteoclasts is realized by cell-cell fusion. In our study,… Show more

“…The dynamic expression changed during osteoblast differentiation and biomineralization of PDLSCs in a temporal manner, suggesting that circRNAs may play a role in osteoblast differentiation and bone formation. Moreover, induction of osteoclast differentiation has recently been associated with change of circRNA profiles 32 . Considering the relatively stable structure, circRNAs can be used as biomarkers for diagnosis and prognosis of bone diseases.…”

“…The procedure of inducing osteoclast different stages during osteoclastogenesis was admitted and described in previous study [16, 39, 40]. In brief, RAW264.7 cells before 3 passages were cultured with osteoclast differentiation medium consisting of DMEM supplemented with 10% FBS, 1% penicillin-streptomycin, 100 ng/ml receptor activator of nuclear factor κB ligand (RANKL; R&D Systems, Minneapolis, MN, USA) and 50 ng/ml macrophage colony stimulating factor (M-CSF; R&D Systems, Minneapolis, MN, USA) for 24 h, 72 h and 96 h to gain Pre-OC, N-OC and M-OC, respectively.…”

“…OCs are specialized multinucleated cells derived from hematopoietic stem cells or monocytes/macrophage progenitor cells [15]. There are three different stages during osteoclastogenesis: preosteoclast (Pre-OC), newborn osteoclast (N-OC) and mature osteoclast (M-OC) [16-18]. EPCs are a population of cells that are found circulating in the bone marrow and peripheral blood, which have the ability to differentiate into mature EC and then participate in vasculogenesis.…”

“…Thus, understanding the characteristics and functions of melanocyte at the molecular and genetic levels may provide crucial information for the development of new regenerative therapies for melanoma. Recently, studies have shown that circular RNAs (circRNAs) have vital roles in a variety of metabolism pathways . Nevertheless, little is known about the functions of circRNAs in the pigmentation pathways, and the role of circRNAs in the melanocyte remains unknown.…”
