“…Our results showed that the genome-wide methylation patterns were similar between two groups. Approximately 9% of all total cytosines and 80% of CG were methylated and only a small proportions of methylation at non-CpG methylation (CHG and CHH) was observed within all regions, which is consistent with previous study (Cai et al, 2020). The high level of DNA methylation in CG content was a specific characteristic of animals.…”

“…A total of 20 wild sub-adult grass carp at the approximately 18-month old determined by annulus characteristics of scale (Cai et al, 2020), were captured using traps and gill nets along the Xijiang river in Zhaoqing City, Guangdong Province, China, within a 1000 m radius from the following coordinates (latitude: 23.1034.8 °N; longitude: 112.4554.0 °E). Subsequently, the fish were transported to breeding base and randomly allocated into two ponds (1.5 m × 1.5 m × 1 m), approximately 250 bp, followed by the blunt-ending, dA addition to 3'-end, finally, adaptor ligation(in this case of methylated adaptors to protect from bisulfite conversion), essentially according to the manufacturer's instructions.…”

“…However, the deferentially methylated genes in Ctenopharyngodon idellus with a high carbohydrate diet were enriched in pathways involved in glucose metabolism, insulin signaling, and lipid metabolism. Most of these obesity and T2DM-involved methylation changes were also observed in mammals (87). Moreover, folic acid (synthetic form of folate) is mainly involved in the methylation processes acting as a methyl donor for the synthetic process of S-adenosylmethionine (108).…”

“…The diet-epigenetic axis is also implicated in pathways related to glucose metabolism and insulin signaling, as demonstrated in animal models. Whole genome DNA methylation analysis of the grass carp, Ctenopharyngodon idellus, revealed no significant changes in methylation level of metabolic genes between different nutritional conditions (high carbohydrate diet/ normal diet) ( 87 ). However, the deferentially methylated genes in Ctenopharyngodon idellus with a high carbohydrate diet were enriched in pathways involved in glucose metabolism, insulin signaling, and lipid metabolism.…”

“…DNMT inhibitors function by blocking the key methylation enzymes such as DNMT1, DNMT3a and DNMT3b, which subsequently blocks DNA methylation. For instance, 5-azacitidine, a structural analogue of cytosine, when incorporated into DNA, is methylated by covalently bound DNMT3a, eventually inhibiting the enzyme activity [ 104 ]. A study by You et al [ 105 ] utilizing insulin-resistant 3T3-L1 adipocytes identified an increased percentage of insulin-stimulated glucose uptake upon treatment with 5-azacitidine [ 105 ].…”
